High mobility group box protein 1 (HMGB1) acts as an endogenous danger molecule that is released from necrotic cells and activated macrophages. We have previously shown that peptide Hp91, whose sequence corresponds to an area within the B-Box domain of HMGB1, activates dendritic cells (DCs) and acts as an adjuvant in vivo. A therapeutic trial using repeated doses of a mouse monoclonal antibody against the tumor-associated antigen (TAA) CO17-1A in metastatic colorectal carcinomas was carried out. Metastatic lesions sampled by repeated thick needle (1.2 mm) biopsies during therapy were examined immunohistochemically for the presence of various TAAs, mouse IgG, complement, and infiltrating leukocytes.

Eight patients with metastasizing colo-rectal carcinoma have been treated with Moab 17-1A. Before infusion the antibodies were incubated in vitro with isolated autologous blood mononuclear cells (AMC) enriched for monocytes/macrophages. Treatment was given in repeated courses (2-4 times) up to a maximum amount of 1000 mg Moab 17-1A.

Using an experimental model of the arterial circulation system, inflow of blood into polyethylene catheters following injection of contrast medium and isotonic saline solution was evaluated. The experiments included an analysis of the influence of density and viscosity of the flush solutions as well as the effect of catheters of different sizes with and without sideholes.

Inflow of blood into intraarterial catheters in dogs following single and continuous injections of saline, contrast medium and polysaccharide fluids were analysed with special reference to the influence of their density and viscosity. Clot formation was also analysed and the favourable properties of one variant of the polysaccharide fluids were demonstrated.
