Several hydroxysteroid dehydrogenase 17-beta 13 variants have previously been identified as protective against metabolic dysfunction-associated steatohepatitis (MASH) fibrosis, ballooning and inflammation, and as such this target holds significant therapeutic potential. However, over 5 years later, the function of 17B-HSD13 remains unknown. Structure-aided design enables the development of potent and selective sulfonamide-based 17B-HSD13 inhibitors.

Post-Acute Sequelae of SARS-CoV-2 infection (PASC), also known as Long-COVID, encompasses a variety of complex and varied outcomes following COVID-19 infection that are still poorly understood. We clustered over 600 million condition diagnoses from 14 million patients available through the National COVID Cohort Collaborative (N3C), generating hundreds of highly detailed clinical phenotypes. Assessing patient clinical trajectories using these clusters allowed us to identify individual conditions and phenotypes strongly increased after acute infection.

Article Synopsis
  • Ontologies are key for managing consensus knowledge in areas like biomedical, environmental, and food sciences, but creating and maintaining them requires significant resources and collaboration among experts.
  • The Dynamic Retrieval Augmented Generation of Ontologies using AI (DRAGON-AI) leverages Large Language Models and Retrieval Augmented Generation to automate the generation of ontology components, showing high precision in relationship creation and ability to produce acceptable definitions.
  • While DRAGON-AI can significantly support ontology development, expert curators remain essential for overseeing the quality and accuracy of the generated content.

Brain magnetic resonance imaging (MRI) of infants with congenital heart disease (CHD) shows brain immaturity assessed via a cortical-based semi-quantitative score. Our primary aim was to develop an infant paralimbic-related subcortical-based semi-quantitative dysmaturation score, termed brain dysplasia score (BDS), to detect abnormalities in CHD infants compared to healthy controls and secondarily to predict clinical outcomes. We also validated our BDS in a preclinical mouse model of hypoplastic left heart syndrome.

Article Synopsis
  • Management decisions in wildlife conservation should consider both habitat selection and demographic performance, as habitat selection alone may not reflect true species viability.
  • This study focuses on the greater sage-grouse to illustrate how mapping habitat selection against survival rates can reveal important mismatches and trade-offs throughout different reproductive life stages.
  • By integrating demographic measures into habitat management, conservation efforts can be more effectively tailored to enhance species survival and resource allocation, particularly during critical life stages.
