Sequencing-based genetic tests have uncovered a vast array of BRCA2 sequence variants. Owing to limited clinical, familial and epidemiological data, thousands of variants are considered to be variants of uncertain significance (VUS). Here we have utilized CRISPR-Cas9-based saturation genome editing in a humanized mouse embryonic stem cell line to determine the functional effect of VUS.

The structural plasticity of proteins at the molecular level is largely dictated by backbone torsion angles, which play a critical role in ligand recognition and binding. To establish the anion-induced cooperative arrangement of the main-chain (mc) torsion, herein, we analyzed a set of naturally occurring CαNN motifs as "static models" for their anion-binding competence through docking and molecular dynamics simulations and decoded its torsion angle influenced mc-driven anion recognition potential. By comparing a pool of 20 distinct sets of CαNN motif with identical sequences in their "anion bound/present, aP" and "anion free/absent, aA" versions, we could discern that there exists a positive correlation between the "difference of anion residence time (ΔR)" and "difference among the main-chain torsion angle" of the aP and aA population.

A cross-sectional study was conducted among the peri-menopausal women to find out determinants of body composition. A total of 200 peri-menopusal women were recruited (≥ 40 years) from rural and urban areas of the district Bilaspur (CG). Among the studied respondents the mean age for peri-menopausal women was 42.

Article Synopsis
  • This review focuses on genetic mutations in kinases related to Parkinson's Disease and analyzes both existing treatments and potential new therapeutic targets.
  • The study highlights four key kinases—PINK1, LRRK2, GAK, and PRKRA—emphasizing that LRRK2 has the most marketed inhibitors, while PINK1, GAK, and PRKRA remain largely unexplored.
  • It calls for increased research on these underinvestigated kinases to develop new therapies that could improve treatment options and address the progression of Parkinson's Disease.

Article Synopsis
  • The study investigates the toxicity of salt on obesity using Drosophila larvae and adult flies, testing varying salt concentrations (50-800 µM).
  • At lower concentrations (50 and 200 µM), significant weight gain and increased feeding behavior were observed, with larvae and adult flies displaying abnormal responses to temperature and light stimuli.
  • The findings reveal that while lower salt concentrations induce stronger adverse effects, higher concentrations deter feeding, suggesting a complex relationship between salt intake and obesity-related behaviors.
