Objective: The aim of the study was to determine whether the clinical outcome of double-blind, placebo-controlled food challenges of patients with atopic dermatitis would be associated with changes in lymphocyte functions.

Methods: Peripheral blood mononuclear cells were prepared from 19 children with atopic dermatitis and stimulated in vitro with the suspected allergen (cow's milk, hen's egg), tetanus toxoid, and pokeweed mitogen. After 14 days in culture, quantitative and qualitative distribution of cell surface marker expression was assessed by flow cytometry, and results were compared with the clinical outcome of a subsequent oral food challenge.

The role of CD45RA T cells on allergen-dependent lymphocyte functions was analyzed in atopic patients. As compared with age-matched nonatopic controls, atopic patients exhibited a significantly (p < 0.01) increased frequency of CD45RA T cells in peripheral blood.

Allergic sensitization is controlled by CD4+ T cells. A complex interaction between antigen-presenting cells, T- and B-cells results in the production of (allergen) specific IgE. Analysis of the lymphokine profile of lymphocytes from patients with bronchialasthma and atopic dermatitis revealed an imbalance in cytokine production.

In order to study cell activation in peripheral blood on bronchial allergen provocation up to 24 h, we investigated 32 asthmatic children, sensitive to house-dust mites. Six healthy young adult volunteers served as controls. Lymphocyte subsets (CD3, CD19, CD4, CD8) and activation markers (CD25-T, HLADR-T, CD23) in peripheral blood as well as soluble IL2-R and soluble ICAM-1 in serum were evaluated.
