Article Synopsis
  • A phase 3 trial analyzed the impact of adding autologous stem-cell transplantation (ASCT) to a treatment regimen involving lenalidomide, bortezomib, and dexamethasone (known as RVD) in patients with newly diagnosed multiple myeloma.
  • Results showed that the group receiving RVD plus ASCT had a median progression-free survival of 67.5 months, compared to 46.2 months for those receiving just RVD, indicating a significantly lower risk of disease progression or death with ASCT.
  • Although progression-free survival improved with ASCT, there was no overall survival advantage, with 5-year survival rates being comparable between the two

The objective of this study is to examine the association between neighborhood socioeconomic status (nSES) and baseline allostatic load (AL) and clinical trial endpoints in patients enrolled in the E1A11 therapeutic trial in multiple myeloma (MM). Study endpoints were symptom burden (pain, fatigue, and bother) at baseline and 5.5 months, non-completion of induction therapy, overall survival (OS) and progression-free survival (PFS).

Background: Bortezomib, lenalidomide, and dexamethasone (VRd) is a standard therapy for newly diagnosed multiple myeloma. Carfilzomib, a next-generation proteasome inhibitor, in combination with lenalidomide and dexamethasone (KRd), has shown promising efficacy in phase 2 trials and might improve outcomes compared with VRd. We aimed to assess whether the KRd regimen is superior to the VRd regimen in the treatment of newly diagnosed multiple myeloma in patients who were not being considered for immediate autologous stem-cell transplantation (ASCT).

Purpose: Observation is the current standard of care for smoldering multiple myeloma. We hypothesized that early intervention with lenalidomide could delay progression to symptomatic multiple myeloma.

Methods: We conducted a randomized trial that assessed the efficacy of single-agent lenalidomide compared with observation in patients with intermediate- or high-risk smoldering multiple myeloma.
