Lenvatinib is an orally effective tyrosine kinase inhibitor used to treat several types of tumors, including progressive, radioiodine-refractory differentiated thyroid cancer and advanced renal cell carcinoma. Although this drug is increasingly used in therapy, its metabolism and effects on the organism are still not described in detail. The main site of attack of these -nucleophiles was predicted using simple orientational rules of aromatic nucleophilic substitution. Thereafter, a series of presumptive 4-ABP metabolites and adducts with cysteine were synthesized, namely, -(4-amino-3-biphenyl)cysteine (ABPC), acetyl--(4-amino-3-biphenyl)cysteine (4-amino-3-biphenylmercapturic acid, ABPMA), -(4-acetamido-3-biphenyl)cysteine (AcABPC), and acetyl--(4-acetamido-3-biphenyl)cysteine (4-acetamido-3-biphenylmercapturic acid, AcABPMA).
