Article Synopsis
  • GRN mutations lead to a condition known as FTD-GRN, which causes frontotemporal dementia; PR006 is a new gene therapy aimed at delivering the granulin gene using an adeno-associated virus.
  • In initial studies, PR006 showed effectiveness in improving various pathological conditions related to FTD-GRN in animal models and was generally well tolerated in non-human primates.
  • An ongoing human trial has reported that PR006 was safe for administration with some transient increases in progranulin levels in cerebrospinal fluid, although some patients experienced treatment-related adverse events, such as CSF pleocytosis and deep vein thrombosis.

Background: The possibility that Alzheimer's disease and related dementias (ADRD) is a modern disease arises from the minimal mention of advanced cognitive decline by ancient Greeks and Romans, who were mainly concerned with the physical frailties of older ages.

Objective: Because standard medical histories of elderly health lacked mention of cognitive decline, we examined texts by Greek and Roman authors that mentioned memory loss and dementia.

Methods: Primary texts of Greco-Roman authors, 8th century BCE into the 3rd century CE, that mentioned cognitive decline were identified and critically evaluated.

Transcatheter aortic valve replacement (TAVR) has become increasingly common as the indications expanded to include valve-in-valve (ViV) applications and a wider patient population with lower surgical risk. Intra-operative coronary arterial occlusion remains a significant source of morbidity, particularly in ViV applications or cases with high-risk anatomy. We present a novel technique for coronary artery protection utilizing a guide extension catheter to secure coronary access during valve deployment and a ViV case demonstration in a patient with prior surgical aortic valve replacement.
