Introduction: Human and preclinical studies have highlighted eNAMPT (extracellular nicotinamide phosphoribosyl transferase) as a druggable TLR4 ligand and DAMP involved in the pathobiology of diverse inflammatory, fibrotic and cancer disorders. This Phase 1 study assesses the safety, pharmacokinetics (PK) and pharmacodynamics (PD) of the humanized eNAMPT-neutralizing ALT-100 mAb as a strategy to address the unmet need for effective anti-inflammatory, anti-fibrotic therapeutics.
Materials And Methods: Healthy male and female volunteers received a single intravenous ALT-100 dose (0.
Lipedema is a chronic condition characterized by abnormal deposition of subcutaneous adipose tissue, leading to pain. The lack of internationally recognized diagnostic criteria complicates the characterization of pain. Physiological parameters such as pain pressure threshold (PPT) represent promising prognostic markers for diagnosing lipedema, yet they remain understudied.

Methods: A retrospective analysis was performed for patients treated at two medical centers (A.O.
Primary myelofibrosis (PMF) is a myeloproliferative neoplasm characterized by a chronic inflammatory state that plays a relevant role in the disease pathogenesis (as proven by high levels of inflammatory cytokines with prognostic significance and by a persistent oxidative stress) and by extensive neoangiogenesis in bone marrow (BM) and spleen. Myeloid-derived suppressor cells (MDSCs) are immature cells that expand in patients with cancer, sepsis or chronic inflammation, favoring tumor onset and progression mainly through the decrease in immune surveillance and the promotion of neoangiogenesis. In this paper, we evaluated the presence of circulating MDSCs in PMF patients, the plasmatic factors involved in their mobilization/expansion and the correlations with laboratory, genetic and clinical parameters.
