Many bioanalytical methods for antisense oligonucleotides (ASOs) using LC-MS have been reported. However, no data have been available on the reproducibility and robustness of a single bioanalytical method for ASOs. As such, in the current study, we evaluated the reproducibility and robustness of LC-MS-based bioanalytical methods for ASOs in multiple laboratories.

Article Synopsis
  • - The study focuses on optimizing an ion-pairing reverse-phase liquid chromatography (IP-LC) method paired with high-resolution mass spectrometry (HRMS) for analyzing oligonucleotide drugs, estimating high sensitivity and selectivity.
  • - Researchers developed a bioanalytical method using mipomersen, a clinically approved treatment, as a model, extracting it from rat plasma and quantifying it with a calibration range of 0.5-250.0 ng/ml.
  • - The new method was validated and found to meet regulatory standards for accuracy, precision, and reliability, indicating its usefulness for measuring mipomersen and potentially other antisense oligonucleotides.
