Global seawater temperatures are increasing and becoming more variable, with consequences for all marine animals including those in food production systems. In several countries around the world,arming of Atlantic salmon (Salmo salar) occurs towards the upper end of the thermal tolerance window for this species, and marked effects on salmon production during summers have been experienced but never empirically investigated. This project tracked the effects of an extreme summer heatwave on two different cohorts of fish stocked into farm cages either during early winter (EW) or late winter (LW).

Peaks in epizootics of sea lice (mostly Caligus chiastos) and blood flukes (Cardicola forsteri) among Southern Bluefin tuna (Thunnus maccoyii) appear to coincide with the onset of an increased mortality. The mortality event occurs 6-12 weeks after T. maccoyii have been transferred into static ranching pontoons from the wild.

The effects of aquaculture related stressors on circulating levels of GH, IGF-I and for the first time, IGF-II in Atlantic salmon (Salmo salar) and rainbow trout (Oncorhynchus mykiss) were investigated. Specifically, circulating growth factor levels were measured in four different experiments. Two 24 h confinement stressor procedures, (one with Atlantic salmon, the other with rainbow trout); following a hypo-osmotic stressor (freshwater bath) in salt water acclimated, adult, Atlantic salmon; and during a 22 day starvation and re-feeding protocol with juvenile Atlantic salmon.

This paper describes the development and validation of a competitive, polyclonal antibody enzyme-immunoassay (EIA) for the measurement of salmon and trout insulin-like growth factor-II (IGF-II). A polyclonal antiserum was raised against a synthetic peptide epitope, corresponding to amino acid residues 1-9 of the N-terminus of mature Atlantic salmon (Salmo salar) IGF-II. The antiserum was purified by hydrophobic charge induction chromatography (HCIC).

The insulin-like growth factors, IGF-I and IGF-II, are single chain polypeptides, which are structurally related to proinsulin and promote proliferation and differentiation of cells in many vertebrate species. Previous attempts to produce recombinant salmon IGF-II (rsIGF-II) were compromised by low expression levels and co-purification of incorrectly cleaved protein with the authentic recombinant product. In this study, a gene containing the coding region for Atlantic salmon (Salmo salar) IGF-II was cloned into a modified pET32a expression vector and transformed into Escherichia coli BL21 trxB (DE3) cells.
