Objectives: Enamel formation is a vulnerable developmental process, susceptible to environmental influences such as excessive systemic fluoride (F) exposure and infant/childhood disease. Methods: A systematic search of electronic databases utilizing medical subject headings (MeSH terms) supplemented by screening of review articles and other relevant texts was undertaken.
Results: Seventy-three articles were included (43 for periodontitis, 30 for caries).
Background: Neuromodulatory medications (NMs), such as amitriptyline, carbamazepine and gabapentin, are used as topical preparations for the management of neuropathic orofacial pain (NOP) and have produced promising preliminary results. The aim of this study was to investigate the effects of three aforementioned NMs on cell lines relevant to the orofacial tissues in vitro as no published studies have examined the effect of these topical NMs.
Methods: Cellular viability was measured using alamarBlue , testing cumulative and specific time point effects of NMs on human skin keratinocytes and oral keratinocytes.

Many genes with crucial roles in zinc homeostasis in mammals respond to fluctuating zinc supply through unknown mechanisms, and uncovering these mechanisms is essential to understanding the process at cellular and systemic levels. We detected zinc-dependent binding of a zinc-induced protein to a specific sequence, the zinc transcriptional regulatory element (ZTRE), in the SLC30A5 (zinc transporter ZnT5) promoter and showed that substitution of the ZTRE abrogated the repression of a reporter gene in response to zinc. We identified the ZTRE in other genes, including (through an unbiased search) the CBWD genes and (through targeted analysis) in multiple members of the SLC30 family, including SLC30A10, which is repressed by zinc.

Principal Findings: Two splice variants of the human SLC30A5 Zn transporter gene (ZnT5) have been reported in the literature. These variants differ at their N- and C-terminal regions, corresponding with the use of different 5' and 3' exons.
The ability to control intracellular levels of transition metals such as Mn(2+), Fe(2+) and Zn(2+) is critical for the virulence of many pathogenic bacteria. In this issue of Molecular Microbiology, Rosch et al. describe the first identification of a Mn(2+) efflux system in bacteria, MntE of Streptococcus pneumoniae, and demonstrate that it is required for virulence in an animal model.
