Synaptic vesicles from rat brain were immobilized on glass microspheres covalently coated with poly-L-lysine. Using a potassium tartrate perfusion medium, the vesicular accumulation and methamphetamine-induced release of (L)-3H-norephinephrine could be conveniently monitored in a flow experiment.

Although 69 patients were initially randomized, 64 patients (34 study, 30 control) are the basis of this report: 5 patients were omitted because large pelvic lymph nodes positive for tumor were found at laparotomy and radical hysterectomy was abandoned. The two study groups were similar in both preoperative and operative risk factors.

The metabolism and disposition of 5-propyl-2'-deoxyuridine (Pr-dUrd) in herpes simplex virus type 1 infections were investigated in cell culture using [14C]Pr-dUrd, [32P]orthophosphate, and several methods including high pressure liquid chromatography and isopycnic centrifugation. Results in infected cells indicate Pr-dUrd 1) is taken up and phosphorylated to mono-, di-, and triphosphates; 2) is incorporated into DNA; 3) preferentially inhibits synthesis of viral DNA; 4) blocks re-initiation of viral DNA synthesis even after removal of the nucleoside from the culture; and 5) exerts these effects early in the course of infection (before 6 h postinfection). Pr-dUrd was not phosphorylated in uninfected cells, and had little or no effect on apparent cellular DNA synthesis in infected or uninfected cells.

Of the analogs tested, E-5-propenyl- and E-5-(2-bromovinyl)-dUTP were more potent inhibitors than dTTP of the enzymes from virus-infected cells, whereas only the former compound showed this effect on the uninfected HeLa enzyme.

Methionine-enkephalin (ME), leucine-enkephalin (LE), phe-met-arg-phe amide (FMRFamide), D-ala2, N-methyl-phe4, met (O)5-ol-enkephalin (FK 33-834), methionine-enkephalin arg6 arg7 (ME arg6 arg7) and beta-endorphin had no effect on basal beating rate of isolated atria at all concentrations up to 10(-5) M. The positive chronotropic effect of norepinephrine (NE) on atrial rate is, however, significantly attenuated by enkephalin peptides.

The effect of acute (10-75 min) exposure of isolated spontaneously beating rat atria to L-triiodothyronine (T3)(12.9-1290 nM) on the positive chronotropic response to (-)-norepinephrine (NE) 10(-10)-10(-4)M) was examined. Preincubation (50 min) with 129 nM T3 increased the maximal response to (-)-NE from 128 +/- 10 to 176 +/- 11 beats/min.

The concentration of drug required to give a 2-log reduction in virus titer was 5 microM for HSV-1 and 23 microM for HSV-2. The anti-HSV-1 activity of this agent was more potent than 5-propyl-dUrd, equivalent to E-5(3,3,3-trifluoropropenyl)-dUrd, and less potent than E-5-bromovinyl-dUrd.

The compunds also were studied for indirect adrenergic agonist activity in the same test system. A definite stereochemical correlation was demonstrated in each case, the exo-isomers being considerably more active than their endo-counterparts both in potentiation and in indirect activity.

Together with these, two isoprenoid alcohols, phytol and dihydrophytol, were identified in anhydrous milk fat. Their identities were established on the basis of chromatographic and mass spectral data and confirmed by comparison with authentic materials.
