Methicillin-resistant Staphylococcus aureus (MRSA) is a major pathogen responsible for serious infections in humans. The overuse of antibiotics has led to the evolution of resistance genes in bacteria. This study aimed to develop a pH-responsive micelle, loaded with therapy drugs and modified with antimicrobial peptides, to treat drug-resistant bacterial infections at varying depths.

Background: Doxorubicin (DOX), a widely used anthracycline chemotherapy agent, is effective against various malignant tumors. However, its clinical application is significantly limited due to dose-dependent cardiotoxicity. Linggui Zhugan Decoction (LGZGD), a traditional Chinese medicine formulation, has demonstrated notable cardioprotective effects.

Rheumatoid arthritis (RA) is an autoimmune disease whose pathophysiology is closely related to inflammation-associated cells and the microenvironment of inflamed joints. This study aimed to develop dual-targeted, reactive oxygen species (ROS)/pH dual-responsive, size-shrinkable intelligent micelles targeting M1 macrophages and fibroblast-like synoviocytes (FLSs) to enhance drug efficacy and safety. These micelles were surface-modified with PEG to prolong their circulation time in the bloodstream and hide the targeting molecules.

Background: Ovarian cancer is difficult to detect in its early stages, and it has a high potential for invasion and metastasis, along with a high rate of recurrence. These factors contribute to the poor prognosis and reduced survival times for patients with this disease. The effectiveness of conventional chemoradiotherapy remains limited.

Ovarian cancer, a highly lethal form of gynecological cancer globally, has witnessed notable advancements in its treatment through the integration of nanotechnology and immunotherapy. Here, we designed a novel astragalus polysaccharide vector (PDA), encapsulating podophyllotoxin (PPT), and modifying methotrexate (DSPE-PEG-MTX) on its surface for targeting ovarian cancer cells with high folate receptor expression. We prepared novel MTX-modified PPT-loaded astragalus polysaccharide micelles (MTX-PPT-micelles) by dialysis method and evaluated their characterization, stability, safety and targeting ability.

Article Synopsis
  • Oxidative stress and neuroinflammation in the aging brain are linked to neurodegenerative diseases like Alzheimer's disease, making effective treatment challenging due to the blood-brain barrier (BBB).
  • A study developed menthol-modified quercetin liposomes (Men-Qu-Lips) to enhance BBB permeability and improve quercetin delivery to the brain.
  • Experimental results showed that Men-Qu-Lips effectively crossed the BBB, reduced oxidative stress and neuroinflammation, protected neurons, and improved learning and memory in aged mice.

Objective: Glioma is the most common and deadly primary malignant tumor in adults. Treatment outcomes are ungratified due to the presence of blood-brain barrier (BBB), glioma stem cells (GSCs) and multidrug resistance (MDR). Docetaxel (DTX) is considered as a potential drug for the treatment of brain tumor, but its effectiveness is limited by its low bioavailability and drug resistance.

Objective: Alzheimer's disease (AD) is a neurodegenerative disease that is associated with aging and is influenced by both genetic and environmental factors. Several studies and clinical trials have demonstrated that resveratrol (Res) and salidroside (Sal) are not only biologically safe but also influence AD biomarker trajectories. However, their clinical applications have been quite limited due to poor specificity, low solubility, and insufficient blood-brain barrier (BBB) penetration.

The blood-brain barrier (BBB) is a protective barrier for brain safety, but it is also a major obstacle to the delivery of drugs to the cerebral parenchyma such as the hippocampus, hindering the treatment of central nervous system diseases such as Alzheimer's disease (AD). In this work, an anti-AD brain-targeted nanodrug delivery system by co-loading icariin (ICA) and tanshinone IIA (TSIIA) into Aniopep-2-modified long-circulating (Ang2-ICA/TSIIA) liposomes was developed. Low-density lipoprotein receptor-related protein-1 (LRP1) was a receptor overexpressed on the BBB.

Rheumatoid arthritis (RA) is an autoimmune disease characterized by synovial joint hyperplasia, joint inflammation, cartilage erosion and bone destruction. Macrophages play an essential role in the pathogenesis of RA, and folate receptor β (FR-β) is highly expressed on the surface of activated synovial macrophages in RA patients. Triptolide (TP) has anti-inflammatory properties, and it can protect the cartilage matrix, but its clinical application has been limited due to poor solubility, low bioavailability and systemic toxicity.
