Effect of urea concentration of pelvic fluid on renal concentrating ability.

The role of the pelvic urine in the renal concentration process was examined in anesthetized antidiuretic rats. The left ureter was cannulated with perfusion and collection catheters so that the composition of the fluid bathing the papilla could be altered by superperfusing solution of known composition without opening the renal pelvis, exposing the papilla, or altering intrapelvic pressure. The effect on urinary osmotic pressure (Uosmol) of papillary superperfusion with solutions of varying urea concentrations and total osmolality was examined.

Replication of DNA by nuclei isolated from soybean suspension cultures.

The isolation procedure involved the preparation of protoplasts, their lysis with a nonionic detergent and purification of nuclei. A method was developed for the analysis of testosterone 17-enanthate 3-benzilic acid hydrazone, 17-beta-estradiol 3,17-dienanthate, and estradiol benzoate combinations dissolved in an oily vehicle. Testosterone 17-enanthate 3-benzilic acid hydrazone was separated from the other drugs and vehicle components by chromatography on an acetonitrile-infusorial earth column followed by quantitation using UV spectroscopy. The estradiol esters were separated from the oil by an additional chromatographic step using a heptane-silanized infusorial earth column prior to quantitation by GLC.

Improved synthesis of n-octyl-beta-D-glucoside: a nonionic detergent of considerable potential in membrane biochemistry.

An improved synthesis for the nonionic detergent n-octyl-beta-D-glucoside (detergent 2) is described. The key modification over earlier methods is the utilization of n-octyl alcohol as the limiting reagent in the condensation between bromide 1 and n-octyl alcohol. A 60-70% overall yield of 2 is obtained without isolation of the intermediate.

Renal tubular site of action of fluoride in Fischer 344 rats.

Methoxyflurane is capable of producing high-output renal failure in some patients and animal models, probably through metabolic liberation of free fluoride. The tubular site of action of fluoride was examined in Fischer 344 rats using clearance techniques. Free water reabsorption (TCH2O) and free water excretion (CH2O) were measured during mannitol or water diuresis in control rats and in rats given methoxyflurane or pretreated with sodium fluoride.

The two cases presented here demonstrate a successful therapeutic modality with resection followed by immediate reconstruction with bone grafts. Fraction C3beta binds Met-tRNAiMet suggesting that a C3beta-Met-tRNAiMet complex may be an intermediate in the formation of the 40 S complex.

Renal tubular transport of 3H-digoxin in saline diuresis in rats.

We evaluated urinary excretion and tubular transport of 3H-digoxin by three different methods in anesthetized rats made diuretic by infusion of 2.5% saline. In one group small volumes of 3H-digoxin and 14C-inulin were injected simultaneously into surface proximal convolutions, and urine was collected serially from both ureters.
