Homozygous MTAP deletion occurs in ~15% of cancers, making them vulnerable to decreases in the concentration of S-adenosylmethionine (SAM). AG-270/S095033 is an oral, potent, reversible inhibitor of methionine adenosyltransferase 2 A (MAT2A), the enzyme primarily responsible for the synthesis of SAM. We report results from the first-in-human, phase 1 trial of AG-270/S095033 as monotherapy in patients with advanced malignancies (ClinicalTrials.

Next-generation data networks need to support Tb/s rates. In-phase and quadrature (IQ) modulation combine phase and intensity information to increase the density of encoded data, reduce overall power consumption by minimizing the number of channels, and increase noise tolerance. To reduce errors when decoding the received signal, intersymbol interference must be minimized.

Article Synopsis
  • The study investigated how acute ischemic stroke (AIS) affects patients with nonvalvular atrial fibrillation (NVAF) who are using oral anticoagulants (OA), specifically comparing direct oral anticoagulants (DOAC) and vitamin K antagonists (VKA).
  • A total of 169 patients were examined, revealing that those on VKA had higher in-hospital mortality rates and poor outcomes 90 days after the stroke compared to DOAC users.
  • The researchers found that switching anticoagulants did not impact stroke recurrence rates, suggesting that further extensive research is needed to optimize treatment for AIS in OA patients.

Objectives: Mental fatigue (MF) can impair cognitive and physical performance in sport. We tested the hypothesis that a shorter adaptive Time Load Dual Back (TLDB) task induces MF faster than a longer Stroop; and subsequently impairs cognitive and intermittent running performance.

Design: This study employed a randomized within-participant design.

Background: The aberrant amplification of mammary luminal progenitors is at the origin of basal-like breast cancers associated with BRCA1 mutations. Integrins mediate cell-matrix adhesion and transmit mechanical and chemical signals that drive epithelial stem cell functions and regulate tumor progression, metastatic reactivation, and resistance to targeted therapies. Consistently, we have recently shown that laminin-binding integrins are essential for the expansion and differentiation of mammary luminal progenitors in physiological conditions.

Ensuring the longevity of wooden constructions depends heavily on the preservation process. However, several traditional preservation methods involving fossil-based compounds have become outdated because they pose a significant risk to the environment and to human health. Therefore, the use of bio-based and bioactive solutions, such as essential oils, has emerged as a more sustainable alternative in protecting wood from biotic attacks.

Purpose: Severe eosinophilic asthma (SEA) patients often present overlapping inflammatory features rendering them eligible for multiple biologic therapies; switching biologic treatment is a strategy adopted to optimize asthma control when patients show partial or no response to previous biologics.

Patients And Methods: ANANKE is a retrospective, multicenter Italian study (NCT04272463). Here, we outline the characteristics and long-term clinical outcomes in naïve-to-biologics and biologics-experienced patients treated with benralizumab for up to 96 weeks.

Introduction/aims: MScanFit MUNE (MScanFit) is a novel tool to derive motor unit number estimates (MUNEs) from compound muscle action potential (CMAP) scans. Few studies have explored its utility in 5q spinal muscular atrophy (SMA5q) patients, assessing only the abductor pollicis brevis (APB) muscle. We aimed to assess different distal muscles in pediatric and adult SMA5q patients, further evaluating clinical-electrophysiological correlations.

This study reports on the two-step manufacturing process of a filtration media obtained by first electrospinning a layer of polycaprolactone (PCL) non-woven fibers onto a paper filter backing and subsequently coating it by electrospraying with a second layer made of pure acidolysis lignin. The manufacturing of pure lignin coatings by solution electrospraying represents a novel development that requires fine control of the underlying electrodynamic processing. The effect of increasing deposition time on the lignin coating was investigated for electrospray time from 2.

  • The study examines the effects of heterozygous mutations in the AFG3L2 and ACO2 genes on the neuro-ophthalmological phenotype of patients with Dominant Optic Atrophy (DOA), comparing them with those having classic OPA1-related DOA.
  • A total of 23 ACO2 and 13 AFG3L2 patients’ eyes were analyzed, revealing that all presented bilateral optic atrophy with varying visual impairment, but ACO2 patients exhibited significantly better retinal thickness metrics than AFG3L2 and OPA1 patients.
  • The findings suggest that while genetic distinctions exist, the clinical presentation of DOA remains largely uniform, with ACO2 mutations linked to better preservation of retinal cells compared

Idebenone, the only approved treatment for Leber hereditary optic neuropathy (LHON), promotes recovery of visual function in up to 50% of patients, but we can neither predict nor understand the non-responders. Idebenone is reduced by the cytosolic NAD(P)H oxidoreductase I (NQO1) and directly shuttles electrons to respiratory complex III, bypassing complex I affected in LHON. We show here that two polymorphic variants drastically reduce NQO1 protein levels when homozygous or compound heterozygous.

The evaluation of pupillary light reflex (PLR) by chromatic pupillometry may provide a unique insight into specific photoreceptor functions. Chromatic pupillometry refers to evaluating PLR to different wavelengths and intensities of light in order to differentiate outer/inner retinal photoreceptor contributions to the PLR. Different protocols have been tested and are now established to assess PLR contribution mediated by melanopsin retinal ganglion cells (mRGCs).
