Background: Despite decades of engineering efforts, recombinant Saccharomyces cerevisiae are still less efficient at converting D-xylose sugar to ethanol compared to the preferred sugar D-glucose. Using GFP-based biosensors reporting for the three main sugar sensing routes, we recently demonstrated that the sensing response to high concentrations of D-xylose is similar to the response seen on low concentrations of D-glucose. The formation of glycolytic intermediates was hypothesized to be a potential cause of this sensing response.

Twenty new compounds, targeting CYP17A1, were synthesized, based on our previous work on a benzimidazole scaffold, and their biological activity evaluated. Granulometric fractionation as a source of additional information on organic-matter and inorganic matrix components of soils using FTIR-photoacoustic spectroscopy (FTIR-PAS) supported by attenuated-total reflection FTIR spectroscopy (ATR-FTIR) for a wide range of aggregate fractions (10-5000 μm) was used to compare the sensitivity, reproducibility, information contents, and representativity of fractionated samples. For chernozem and sod-podzolic soils and different agricultural-use chernozem samples, differences in the composition were found, manifested in normalized spectra of microaggregate fractions, with the range of 10-100 μm bearing the complete information. Most changes are observed in the soil organic matter range (1900-1340 cm), although these changes are slight, and in the soil-matrix region (550-300 cm).

CYP17A1 is a key enzyme in the steroidogenic pathway that produces androgens among other steroids, and it is implicated in prostate cancer. FTIR photoacoustic spectroscopy was used to approach inorganic matrix components and organic-matter constituents of chernozem size fractions (1-5000 μm, by dry sieving) with a different history of use (from intact steppe to permanent bare fallow, a continuous long-term field experiment). The conditions of FTIR photoacoustic measurements in continuous-scan modes were compared with attenuated total reflection measurements, the advantages of photoacoustic measurements resulting from a higher intensity of the incident radiation and signal-generating volume were discussed. Overtone peaks of quartz as a soil matrix component at 2000-1700 cm were selected as a possible internal-standard (guide) bands for the comparison of photoacoustic spectra.

A new methodology for site-selective and stereoselective C-H functionalization of aminocyclopropanes via directed remote lithiation has been developed. Treatment of N-directing group (DG = pivaloyl, tetramethylsuccinimidoyl) arylcyclopropanes with t-BuLi results in a clean β-lithiation and, following quench with electrophiles, leads to a range of cyclopropane derivatives. Sequential double lithiation-methylation to give a dimethylated cyclopropane has been achieved.

The conformational states of the self-complementary deoxyhexanucleotides d(GCATGC) and d(GCTAGC) capable of forming hairpin structures in aqueous solution have been studied by one- and two-dimensional 1H NMR spectroscopy and molecular dynamics simulations. The equilibrium thermodynamic parameters of the formation of duplex and hairpin forms have been determined, and the spatial structures of the d(GCATGC) and d(GCTAGC) conformers have been calculated. The molecular basis of the marked structure-activity relationship for a homologous series of DNA-binding phenoxazone drugs (ActII-ActIV) has been investigated by NMR spectroscopy and molecular mechanics. A stochastic model for the NMR analysis of the heteroassociation of two aromatic compounds was developed, which takes into account all physically possible reactions of association of molecules in solution. Expressions for calculating the experimentally observed proton chemical shift were obtained in the general form, and an algorithm for calculating the parameters of heteroassociation using the stochastic model was proposed. The effects of limitations of the basic and general models, as compared with the stochastic model, on the model parameters of the heteroassociation of various biologically active aromatic molecules was analyzed.

Changes in the functional parameters of the rat heart mitochondria were studied in time after a single intraperitoneal administration of rubomycin, the rubomycin combination with 4-hydroxy-2,2,6,6-tetramethylpiperidine-N-oxyl (TEMPO-OH) and ruboxyl, a nitroxyl derivative of rubomycin. The administration of rubomycin resulted in inhibition of the heart mitochondria bioenergetic functions (a decrease in the respiration control coefficient, RCC, and the respiration rate, RR, on phosphorylation) during respiration in the presence of NAD(+)-dependent substrates 6 to 24 hours after the administration. Later the mitochondria functions recovered while in 2 to 3 weeks a secondary decrease in the RCC and RR was observed.

The impact of daunorubicin, emoksil in sublethal doses and daunorubicin mixtures with a nitroxyl radical of 2,2,6,6-tetramethyl-4-oxopiperidine-1-oxyl (NR) on synthesis of DNA and RNA in some organs of rats was studied. Daunorubicin and emoksil induced marked (by 80 to 90%) inhibition of DNA synthesis in all the examined organs even within the first hours of administration. In the heart, DNA synthesis remained lower by the end of the experiment.
