Efforts to prevent dementia can benefit from precision interventions delivered to the right population at the right time; that is, when the potential to reduce risk is the highest. Young adults (aged 18-39 years) are a neglected population in dementia research and policy making despite being highly exposed to several known modifiable risk factors. The risk and protective factors that have the biggest effect on dementia outcomes in young adulthood, and how these associations differ across regions and groups, still remain unclear.

The negative interference of treatments between the acetylcholinesterase inhibitor rivastigmine and the tau aggregation inhibitor hydromethylthionine mesylate (HMTM) has been reported in Line 1 tau-transgenic mice, which overexpress a truncated species of tau protein that is found in the core of paired helical filaments in Alzheimer´s disease (AD). However, little is known about whether such interactions could affect synapses in mice overexpressing tau carrying pathogenic mutations. Here, we have used Line 66 (L66) mice which overexpress full-length human tau carrying the P301S mutation as a model in which tau accumulates in synapses.

Article Synopsis
  • The study aimed to identify risk factors for fetal alcohol spectrum disorders (FASD) by interviewing mothers of first-grade children, both with FASD diagnoses and controls.
  • Results revealed significant differences in alcohol consumption patterns between mothers of children with FASD and control mothers, with those with FASD generally consuming more alcohol before and during pregnancy.
  • The analysis highlighted maternal factors such as alcohol intake, liver issues, depression, and late prenatal care as key risk indicators for FASD, with less influence from paternal alcohol consumption once maternal drinking was accounted for.

Article Synopsis
  • ALS is a severe neurodegenerative disease characterized by the buildup of misfolded proteins in motor neurons, prompting researchers to find ways to reduce this burden for potential treatment.
  • Antisense oligonucleotides (ASOs) have been identified as a promising option to target proteins like SOD1 that cause mutations, but their delivery to the central nervous system is challenging due to the blood-brain barrier.
  • The study demonstrates that using transcranial focused ultrasound (FUS) along with calcium phosphate lipid nanoparticles significantly enhances the delivery of a SOD1 ASO into the brain of mice, leading to reduced SOD1 levels and improved motor neuron survival without damaging brain tissue.

Background: Historically, word of mouth, total citation count, H-index, and expert opinion were some of the primary metrics used to identify the most influential papers. This method is subject to significant bias. The disruption score was initially created with the intent of measuring the degree to which a publication introduces a new idea, is repeatedly referenced and may be used as an additional publication evaluation metric.

Lifespan is an integrative phenotype whose genetic architecture is likely to highlight multiple processes with high impact on health and aging. Here, we conduct a genetic meta-analysis of longevity in Diversity Outbred (DO) mice that includes 2,444 animals from three independently conducted lifespan studies. We identify six loci that contribute significantly to lifespan independently of diet and drug treatment, one of which also influences lifespan in a sex-dependent manner, as well as an additional locus with a diet-specific effect on lifespan.

Article Synopsis
  • This study uses multi-modal MRI to investigate neurobiological differences between anorexia nervosa (AN) and bulimia nervosa (BN), revealing structural and functional brain changes linked to these eating disorders.
  • Key findings include reduced gray matter volume in specific brain regions (like the orbitofrontal cortex) and decreased cortical thickness, particularly in anorexia patients, which are associated with impulsivity and cognitive restraint regarding eating behaviors.
  • The results suggest that these brain changes affect reward processing and contribute to the persistence of eating disorder symptoms, highlighting potential targets for future treatment interventions.
