Background: The management of advanced prostate cancer continues to evolve rapidly, particularly with the earlier use of survival-prolonging therapies in metastatic castration-sensitive prostate cancer (mCSPC). Though approved prior to the use of intensification therapy in mCSPC, taxane-based chemotherapies remain a relevant option for patients with metastatic castration-resistant prostate cancer (mCRPC). However, there is little evidence determining the outcomes of taxane chemotherapies as the first subsequent taxane (FST) in mCRPC pts who received docetaxel intensification (DI) in mCSPC.

In recent years, the prevalence of infertility has increased, and appears to affect approximately one in six couples. Some of them must perform assisted reproductive techniques (ART) in order to achieve pregnancy. As a result, growing interest has arisen about predictive factors of pregnancy and live birth with and without ART.

TRPV6 calcium channel is a prospective target in prostate cancer (PCa) since it is not expressed in healthy prostate while its expression increases during cancer progression. Despite the role of TRPV6 in PCa cell survival and apoptotic resistance has been already established, no reliable tool to target TRPV6 channel in vivo and thus to reduce tumor burden is known to date. Here we report the generation of mouse monoclonal antibody mAb82 raised against extracellular epitope of the pore region of the channel.

Study Question: What are the potential risk factors for poor oocyte recuperation rate (ORR) and oocyte immaturity after GnRH agonist (GnRHa) ovulation triggering?

Summary Answer: Lower ovarian reserve and LH levels after GnRHa triggering are risk factors of poor ORR. Higher BMI and anti-Müllerian hormone (AMH) levels are risk factors of poor oocyte maturation rate (OMR).

What Is Known Already: The use of GnRHa to trigger ovulation is increasing.

Article Synopsis
  • Women with polycystic ovary syndrome (PCOS) often have high levels of DHEAS, a hormone, and researchers want to learn more about this.
  • A study at Lille University looked at more than 1,200 women with PCOS and compared them with 517 women without it to see how many had increased DHEAS levels.
  • They found that women with PCOS had higher DHEAS levels than those without, especially certain types of PCOS, and DHEAS levels were related to other hormones in the body.

Research Question: Does colorectal endometriosis surgery prior to IVF ± intracytoplasmic sperm injection (ICSI) impact cumulative live birth rates?

Design: This retrospective, monocentric study (Lille University Hospital) was conducted between 1 January 2007 and 31 December 2018. Two groups of patients from the JFIV database were included: a group undergoing IVF±ICSI alone (120 patients, 215 oocyte retrievals), and a group undergoing surgery and then IVF±ICSI (69 patients, 109 oocyte retrievals). The mode of management was decided after a multidisciplinary team meeting.

Objectives: Since 2015, in France, men and women who have never procreated are allowed to donate their gametes. This has led to an increase in the number of female oocyte donors, whereas there are many couples waiting for gametes that have a long waiting time. The aim of this study is to compare the results of donation with oocytes from nulliparous and non-nulliparous donors.

Research Question: How does the typology and effect of pain vary between infertile patients with or without endometriosis during the different stages of the IVF process?

Design: A prospective, monocentric, observational cohort study was conducted at Lille University Hospital between November 2019 and June 2021. The study was proposed to all patients starting an IVF cycle. Pain assessment questionnaires using validated scales (about type of pain, without specific location), were completed by patients at key points during IVF: before starting treatment, at the end of stimulation and on the day of oocyte retrieval.

Article Synopsis
  • The study investigates whether men with specific ultrastructural defects in sperm flagella have lower success rates in intracytoplasmic sperm injection (ICSI) and worse outcomes for pregnancies and newborns.
  • A retrospective analysis of 189 ICSI cycles from men with three types of flagellar defects (DFS, PFD, NSFA) was conducted, comparing their outcomes with a control group.
  • Results showed no significant negative impact of these defects on fertilization, pregnancy, or delivery rates, indicating generally favorable outcomes despite the presence of flagellar abnormalities.

  • Systemic sclerosis (SSc) is an autoimmune disease with few treatment options, making the development of new therapies essential.
  • Researchers focused on galectin-3 (Gal-3), a protein linked to SSc, and found significant transcriptomic markers in patient blood samples that correlate with disease severity and complications.
  • They created monoclonal antibodies that target Gal-3, which showed promise in a mouse model by reducing skin and lung damage and reversing adverse gene expression changes associated with SSc, highlighting potential for new treatment strategies.

Research Question: Clomiphene citrate (CC) is one of the first-line treatments for ovulation induction in women with anovulatory polycystic ovary syndrome (PCOS). However, nearly 1 out of 2 women is resistant to 50 mg/day of CC. The objective of this study is to investigate the clinical, biological, and/or ultrasound factors that may predict the resistance to 50 mg/day of CC in the first cycle of treatment in women with anovulatory PCOS.

Article Synopsis
  • The study investigates whether serum anti-Müllerian hormone (AMH) levels can predict cumulative live birth (CLB) rates in women undergoing assisted reproductive technologies (ART) like IVF/ICSI, or in those trying to conceive naturally, with findings indicating a link after IVF but a lack of data for natural conception.* -
  • Serum AMH levels serve as a marker of ovarian reserve and are useful for predicting ovarian response, though the ability to forecast CLB in natural conception or through IUI remains uncertain.* -
  • A systematic review and meta-analysis of 32 studies revealed a non-linear positive relationship between AMH levels and CLB rates post-IVF/ICSI, but no specific threshold was found that could guarantee live

The association between endometriosis and autoimmune diseases is well known, however no acquired platelet function defect has been described so far. We describe the case of two patients with endometriosis associated with an antiplatelet glycoprotein VI (anti-GPVI) antibody. The two women with deep pelvic endometriosis associated with secondary infertility presented a mild bleeding tendency, a deficient platelet aggregation response to collagen, convulxin or CRP and a severe GPVI deficiency.
