The optimal modalities of kidney replacement therapy (KRT) in the ICU remain debated. Intermittent haemodialysis (IHD) and continuous veno-venous haemofiltration (CVVH) are the two main methods. Intermittent haemodialysis requires a water treatment system, which may not be available in all jurisdictions.

Background: Exposure to community violence is associated with increased occurrence of substance use disorders (SUD). The self-medication hypothesis states that heightened negative emotionality may underlie the link between exposure to community violence and SUD. However, it is not well-understood if access to community resources, a broader public health approach, influences the purported psychological mechanisms underlying the link between community violence exposure and SUD.

Considerable heterogeneity exists in the expression of complex human behaviors across the cognitive, personality and mental health domains. It is increasingly evident that individual variability in behavioral expression is substantially affected by sociodemographic factors that often interact with life experiences. Here, we formally address the urgent need to incorporate intersectional identities in neuroimaging studies of behavior, with a focus on research in mental health.

Background And Aims: Estimating the genetic risk of coronary artery disease (CAD) is now possible by aggregating data from genome-wide association studies (GWAS) into polygenic risk scores (PRS). Combining multiple PRS for specific circulating blood lipids could improve risk prediction. Here, we sought to evaluate the performance of PRS derived from CAD and blood lipids GWAS to predict the incidence of CAD.

Predicting bacteriophage infection of specific bacterial strains promises advancements in phage therapy and microbial ecology. Whether the dynamics of well-established phage-host model systems generalize to the wide diversity of microbes is currently unknown. Here we show that we could accurately predict the outcomes of phage-bacteria interactions at the strain level in natural isolates from the genus Escherichia using only genomic data (area under the receiver operating characteristic curve (AUROC) of 86%).

Unlike mammals, some nonmammalian species recruit Müller glia for retinal regeneration after injury. Identifying the underlying mechanisms may help to foresee regenerative medicine strategies. Using a model of retinitis pigmentosa, we found that Müller cells actively proliferate upon photoreceptor degeneration in old tadpoles but not in younger ones.

Article Synopsis
  • - The study investigates how brain mechanisms related to cocaine use disorder involve both cortical and subcortical systems, emphasizing the importance of large-scale functional brain networks and the dopamine system.
  • - Previous research predominantly focused on cortico-striatal circuits, but this study shifts attention to how functional connectivity patterns are associated with neurotransmitter receptor densities in cocaine users.
  • - Findings reveal that specific patterns of connectivity in the brains of individuals with cocaine use disorder correspond with the spatial densities of dopamine D receptors, suggesting that these receptor distributions may influence brain connectivity associated with substance use.

  • Endotracheal intubation (ETI) is crucial for critically ill patients
  • , but it has risks like hypoxemia; preoxygenation is used to enhance safety but can have unreliable monitoring through standard pulse oximeters (SpO2).
  • The Oxygen Reserve Index (ORI)
  • is proposed as a more reliable continuous measure during preoxygenation, targeting an ORI value of at least 0.6 over one minute instead of the standard SpO2 100% goal to improve outcomes.
  • A trial with 950 critically ill adults
  • will compare ORI monitoring against standard SpO2 monitoring during ETI; outcomes include the lowest SpO2 during intubation and cognitive

Article Synopsis
  • * Genome sequencing revealed that resistant bacteria often mutate the same components (K15 capsule and LPS) in different conditions, indicating predictable resistance pathways.
  • * The researchers found that while some mutants maintained high virulence, others had reduced fitness, suggesting that a variety of phages should be used in therapies to effectively tackle these evolving bacterial strains.

Background: Hypertension is a major source of morbidity and mortality worldwide, particularly for racial and ethnic minorities who face higher rates of hypertension and worse health-related outcomes. Recent research has reported on protective associations between classic psychedelics and hypertension; however, there is a need to explore how race and ethnicity may moderate such associations.

Methods: We used data from the National Survey on Drug Use and Health (2005-2014) to assess whether race and ethnicity moderate the associations between classic psychedelic use - specifically psilocybin - and past year hypertension.

Background: Previous retrospective research has shown that maintaining prone positioning (PP) for an average of 40 h is associated with an increase of survival rates in intubated patients with COVID-19-related acute respiratory distress syndrome (ARDS). This study aims to determine whether a cumulative PP duration of more than 32 h during the first 2 days of intensive care unit (ICU) admission is associated with increased survival compared to a cumulative PP duration of 32 h or less.

Methods: This study is an ancillary analysis from a previous large international observational study involving intubated patients placed in PP in the first 48 h of ICU admission in 149 ICUs across France, Belgium and Switzerland.

Upregulation of soluble tumor necrosis factor (sTNF) cytokine signaling through TNF receptor 1 (TNFR1) and subsequent neuronal hyperexcitability are observed in both animal models and human chronic neuropathic pain (CNP). Previously, we have shown that estrogen modulates sTNF/TNFR1 signaling in CNP, which may contribute to female prevalence of CNP. The estrogen-dependent role of TNFR1-mediated supraspinal neuronal circuitry in CNP remains unknown.

Article Synopsis
  • The study investigates the airway occlusion pressure at 100 ms (P0.1) as an indicator of central respiratory drive and its correlation with patient outcomes in ICUs.
  • It analyzes data from intubated patients in French ICUs, showing that variations in P0.1 are related to factors like dyspnea and respiratory rate, and highlights the distribution of P0.1 values among the patients assessed.
  • The findings indicate that higher P0.1 values correlate with increased 90-day mortality rates and longer mechanical ventilation duration, suggesting that abnormal P0.1 levels may reflect worsening patient conditions.

Background: The biological mechanisms that contribute to cocaine and other substance use disorders involve an array of cortical and subcortical systems. Prior work on the development and maintenance of substance use has largely focused on cortico-striatal circuits, with relatively less attention on alterations within and across large-scale functional brain networks, and associated aspects of the dopamine system. The brain-wide pattern of temporal co-activation between distinct brain regions, referred to as the functional connectome, underpins individual differences in behavior.
