X-Band (9 GHz) and W-band (94 GHz) EPR spectral parameters of the new spin-label in its free form and covalently attached to an amino acid cysteine and a tripeptide glutathione were studied as a function of pH and solvent polarity. The pKa value of the protonatable tertiary amino group of the spin-label was found to be unaffected by other ionizable groups present in side chains of unstructured small peptides.

In some cell types, oscillations in the concentration of free intracellular calcium ([Ca2+]) are accompanied by oscillations in the concentration of inositol 1,4,5-trisphosphate ([IP3]). However, in most cell types it is still an open question as to whether oscillations in [IP3] are necessary for Ca2+ oscillations in vivo, or whether they merely follow passively. Using a wide range of models, we show that the response to an artificially applied pulse of IP3 can be used to distinguish between these two cases.

The method is based on the detection of products of the exchange reaction of thiols with a newly synthesized fluorinated disulfide, 2,3,5,6-tetrafluoro-4-mercaptobenzoic acid (BSSB). A significant broadening of the 19F NMR signal of BSSB in the presence of thiols was observed and attributed to the exchange reaction between the parent disulfide and 2,3,5,6-tetrafluoro-4-mercaptobenzoic acid.

Sulfite anion trioxide (SO(3)(-*)) radical formation was measured in mice, preloaded with the spin trap, 5-(diethoxy-phosphoryl)-5-methyl-pyrrolidine-N-oxide, (DEPMPO). Based on spin trapping NMR, a surprising quantity of reduced SO(3)(-*)-adduct was observed that did not depend on co-administration of oxidizing agents, suggesting a possible nonradical reaction between (bi)sulfite and DEPMPO.

Whole-cell currents in single smooth muscle cells freshly isolated from the rat tail artery have been studied using patch-clamp recordings techniques. Outward current evoked by depolarizing steps from -70 mV consisted of the initial fast and subsequent sustained components. The former was inhibited by 10 mM nifedipine and Ca(2+)-free solution application whereas the latter decreased by only 16% under these conditions.

Spontaneous transient outward currents (STOCs) were studied in the rat tail artery smooth muscle cells using standard patch-clamp recording techniques in the whole-cell configuration. STOCs evoked by membrane depolarization from -30 to 20 mV varied in size from 50 to 1000 pA, their amplitude increased with membrane depolarization. These currents were inhibited by 1 mM TEA+.

Earlier we reported an ESR method of quantitative determination of sulfhydryl groups. The method is based on the application of the imidazoline biradical disulfide label, R1S-SR1, which participates in the reaction of thiol-disulfide exchange followed by dramatic changes in ESR spectra. One of the disadvantages of the application of R1S-SR1 at physiological conditions is the requirement of excess of the biradical compared with thiol content which results in the consumption of the thiols and irreversible damage of the system under study.

It is found that the shift in pKa of the amino group of the imidazolidine radical from its value of 4.6 in water depends both on the interfacial polarity (delta pKapol) and on the electrostatic surface potential (delta pKael) when it is positioned at the bilayer/water interface by an anchoring hydrocarbon tail.
