Laccases are important and valuable enzymes with a great potential for biotechnological applications. In this study, two novel laccases, LacHU1 and LacHU2, from sp. HU have been purified and characterized.

Typical laccases have four copper atoms, which form three different copper centers, of which the T1 copper is responsible for the blue color of the enzyme and gives it a characteristic absorbance around 610 nm. Several laccases have unusual spectral properties and are referred to as yellow or white laccases. Only two yellow laccases from the Ascomycota phylum have been described previously, and only one amino acid sequence of those enzymes is available.

Flavonoids exhibit prooxidant cytotoxicity in mammalian cells due to the formation of free radicals and oxidation products possessing quinone or quinomethide structure. However, it is unclear how the cytotoxicity of flavonoids depends on the ease of their single-electron oxidation in aqueous medium, i.e.

The kinetics of potassium triiodide (KI(3)) formation during fungal laccase action was investigated in presence of methyl syringate (MS). The recombinant forms of Polyporus pinsitus (rPpL), Myceliophthora thermophila (rMtL), Coprinus cinereus (rCcL), and Rhizoctonia solani (rRsL) laccases were used. The triiodide formation rate reached 6.

Article Synopsis
  • Researchers developed biosensors using graphite electrodes (GE) and printed graphite electrodes (PGE) that incorporate recombinant fungal laccase enzymes from two different species (Polyporus pinsitus and Myceliophthora thermophila).
  • The biosensors demonstrated varying levels of sensitivity and calibration responses depending on the enzyme concentration and type, with notable sensitivities ranging from 0.11 to 4 A/M under certain conditions.
  • Both biosensor types showed consistent performance over short durations but experienced reduced sensitivity over extended use, effectively detecting various phenolic compounds.
