There is a pressing need for new cell-laden, printable, biomaterials that are rigid and highly biocompatible. These materials can mimic stiffer tissues such as cartilage, fibrotic tissue and cancer microenvironments, and thus have exciting applications in regenerative medicine, wound healing and cancer research. Self-assembled peptides (SAPs) functionalised with aromatic groups such as Fluorenyl-9-methoxycarbonyl (Fmoc) show promise as components of these biomaterials.

Human DNA ligase 1 (LIG1) finalizes DNA repair pathways by an ultimate ligation step and discriminates against nicks containing unusual ends, yet the contribution of the conserved active site residues for faithful end joining remains unknown. Here, using biochemistry, X-ray crystallography, and single-molecule approaches, we comprehensively characterized LIG1 mutants carrying Ala(A) and Leu(L) substitutions at the active site residues Phe(F)635 and Phe(F)872. Our results showed an abolished ligation of nick DNA substrates with all 12 non-canonical mismatches, while the mutagenic nick sealing of oxidatively damaged ends by wild-type enzyme is significantly reduced by F635A/L and F872A/L substitutions.

Base excision repair (BER) maintains genome integrity by fixing oxidized bases that could be formed when reactive oxygen species attack directly on the DNA. We previously reported the importance of a proper coordination at the downstream steps involving gap filling by DNA polymerase (pol) β and subsequent nick sealing by DNA ligase (LIG) 1 or 3α. Yet, how the fidelity of 8-oxoG bypass by polβ affects the efficiency of ligation remains unclear.

DNA ligase 1 (LIG1) joins Okazaki fragments during the nuclear replication and completes DNA repair pathways by joining 3'-OH and 5'-PO4 ends of nick at the final step. Yet, the mechanism of how LIG1 searches for a nick at single-molecule level is unknown. Here, we combine single-molecule fluorescence microscopy approaches, C-Trap and total internal reflection fluorescence (TIRF), to investigate the dynamics of LIG1-nick DNA binding.

Article Synopsis
  • - Neuro-COVID includes symptoms such as neurological pain, memory loss, and cognitive disruptions that can last for months, classified as Post-Acute Sequelae of COVID-19 (PASC).
  • - Research suggests that protein fragments from the SARS-CoV-2 virus can form amyloid nanofibrils, potentially contributing to these neurological symptoms.
  • - Recent findings indicate that manipulating the environment of these protein fragments can lead to less toxic amyloid structures, impacting neuron function without being as harmful, stressing the need to understand amyloid behavior for addressing neuro-COVID.

Peatland drainage is a large source of anthropogenic CO emissions. While conversion to agriculture is widely acknowledged to lead to "irrecoverable" carbon (C) losses, in contrast the C impacts of peatland forestry are poorly understood, especially in intensively managed plantations. Losses of C from peat oxidation are highly variable and can be compensated for by gains of C in trees, depending on the lifecycle of the timber and timescale considered.

Article Synopsis
  • The CHAMPP4KIDS study looked at how early childhood providers in Canada feel about a universal parenting resource called the Parenting for Lifelong Health tip sheets.
  • Providers thought the tip sheets were helpful and relevant to parents, but they weren't sure about just giving them out without extra support.
  • They suggested making improvements to the tip sheets to be easier to read and more culturally relevant so that more families could benefit from them.

Fullerenes are a class of carbon nanomaterials that find a wide range of applications in biomedical fields, especially for photodynamic cancer therapy because of its photosensitive effect. However, hydrophobic fullerenes can only be dispersed in organic solvents which hinders their biomedical applications. Here, we report a facile method to prepare highly water-dispersible fullerene (C)-polymer nanoparticles with hydrodynamic sizes of 50-70 nm.

Background: Multicomponent interventions with carers of people with dementia demonstrate positive effects on the health and quality of life for carers and care recipients. The World Health Organization's iSupport for Dementia is an evidence-based online psychoeducation programme for carers. However, the programme was mainly implemented as a self-learning tool which might have limited its positive effects on carers and care recipients.

Ribonucleotides in DNA cause several types of genome instability and can be removed by ribonucleotide excision repair (RER) that is finalized by DNA ligase 1 (LIG1). However, the mechanism by which LIG1 discriminates the RER intermediate containing a 5'-RNA-DNA lesion generated by RNase H2-mediated cleavage of ribonucleotides at atomic resolution remains unknown. Here, we determine X-ray structures of LIG1/5'-rG:C at the initial step of ligation where AMP is bound to the active site of the ligase and uncover a large conformational change downstream the nick resulting in a shift at Arg(R)871 residue in the Adenylation domain of the ligase.
