A pivotal animal model for development of anticancer molecules is mice with subcutaneous tumors, grown by injection of xenografted tumor cells, where micro-Computed Tomography (µCT) of the mice is used to analyze the efficacy of the anticancer molecule. Manual delineation of the tumor region is necessary for the analysis, which is time-consuming and inconsistent, highlighting the need for automatic segmentation (AS) tools. This study introduces a preclinical µCT database, comprising 452 whole-body scans from 223 individual mice with subcutaneous tumors, spanning ten diverse µCT datasets conducted between 2014 and 2020 on a preclinical PET/CT scanner, making it the hitherto largest dataset of its kind.

Article Synopsis
  • The study evaluated the length of the Achilles tendon and any impact on movement after non-surgical treatment of acute Achilles tendon rupture, with follow-up 4-5 years post-injury.
  • Results showed that the injured tendon was 1.7 cm longer than the healthy tendon, but overall walking mechanics remained symmetrical despite a slight delay in heel lift on the injured side.
  • Changes were noted in calf circumference, Achilles tendon resting angle, and passive ankle dorsiflexion on the injured leg, but no significant differences were found in foot pressure distribution patterns.

Background: In this study we have evaluated the accuracy of automatic, deformable structure propagation from planning CT and MR scans for daily online plan adaptation for MR linac (MRL) treatment, which is an important element to minimize re-planning time and reduce the risk of misrepresenting the target due to this time pressure.

Methods: For 12 high-risk prostate cancer patients treated to the prostate and pelvic lymph nodes, target structures and organs at risk were delineated on both planning MR and CT scans and propagated using deformable registration to three T2 weighted MR scans acquired during the treatment course. Generated structures were evaluated against manual delineations on the repeated scans using intra-observer variation obtained on the planning MR as ground truth.

Background And Purpose: In radiotherapy, treatment response is generally evaluated many weeks after end of the treatment course. If the treatment outcome could be predicted during radiotherapy better tumour control could be achieved through timely adaptation of the treatment strategy. In this study intra-treatment change based on the diffusion MRI outlined viable tumour volume (VTV) was assessed and compared to the standard GTV to study their outcome prediction capacity.

Assessing target occupancy is critical for establishing proof-of-mechanism for novel inhibitors and to determine whether robust target inhibition can be achieved at tolerated doses. This is challenging in the clinic using conventional methods due to the need for untreated controls. We describe a new mass spectrometry approach to quantitatively assess target occupancy for covalent inhibitors that does not require untreated controls, and apply the method to the KRAS inhibitor ARS-1620.

The indigenous people of Greenland, the Inuit, have lived for a long time in the extreme conditions of the Arctic, including low annual temperatures, and with a specialized diet rich in protein and fatty acids, particularly omega-3 polyunsaturated fatty acids (PUFAs). A scan of Inuit genomes for signatures of adaptation revealed signals at several loci, with the strongest signal located in a cluster of fatty acid desaturases that determine PUFA levels. The selected alleles are associated with multiple metabolic and anthropometric phenotypes and have large effect sizes for weight and height, with the effect on height replicated in Europeans.

Because of past limitations in samples and genotyping technologies, important questions about the history of the present-day Greenlandic population remain unanswered. In an effort to answer these questions and in general investigate the genetic history of the Greenlandic population, we analyzed ∼200,000 SNPs from more than 10% of the adult Greenlandic population (n = 4,674). We found that recent gene flow from Europe has had a substantial impact on the population: more than 80% of the Greenlanders have some European ancestry (on average ∼25% of their genome).

Aims: This study evaluates the agreement between echocardiographic and cardiac magnetic resonance (CMR) imaging data, and the impact a discrepancy between the two may have on the clinical diagnosis of arrhythmogenic right ventricular cardiomyopathy (ARVC).

Methods And Results: From the Nordic ARVC Registry, 102 patients with definite ARVC who had undergone both echocardiography and CMR were included (median age 42 ± 16 years, 36% female, 78% probands). Patients were divided into two groups according to CMR-positive or -negative criteria, and the echocardiographic data were compared between the two.

Purpose: Conventional ultrasound methods for acquiring color flow images of the blood motion are limited by a relatively low frame rate and are restricted to only giving velocity estimates along the ultrasound beam direction. To circumvent these limitations, the Plane Wave Excitation (PWE) method has been proposed.

Material And Methods: The PWE method can estimate the 2D vector velocity of the blood with a high frame rate.

Only a fraction of Helicobacter pylori (HP)-infected individuals develop clinical disease. Recent research indicates that immunological mechanisms may be important for understanding the pathophysiology of HP infection. Differences in the individual cellular immune response may reflect the clinical diversity.

Acyl-migrated isomers of drug beta-1-O-acyl glucuronides have been implicated in drug toxicity because they can bind to proteins. The acyl migration and hydrolysis of S-naproxen-beta-1-O-acyl glucuronide (S-nap-g) was followed by dynamic stopped-flow HPLC-1H NMR and HPLC methods. Nine first order rate constants in the chemical equilibrium between six species (S-nap-g, its alpha/beta-2-O-acyl, alpha/beta-3-O-acyl, alpha/beta-4-O-acyl, and alpha-1-O-acyl-migration isomers, and S-naproxen aglycone) were determined by HPLC-UV studies in 25 mM potassium phosphate buffer, pH 7.
