Dietary restriction (DR) has long been recognized as a powerful intervention for extending lifespan and improving metabolic health across species. In laboratory animals, DR-typically a 30%-40% reduction in caloric intake-delays aging and enhances mitochondrial function, oxidative defense, and anti-inflammatory pathways. In humans, findings from the CALERIE™ trial confirm DR's potential benefits, with a 25% caloric reduction over 2 years resulting in reduced visceral fat, improved cardiometabolic health, and favorable gene expression changes linked to proteostasis, DNA repair, and inflammation.

Article Synopsis
  • The study investigated the impact of replacing fish oil with olive oil in sea bass diets, both with and without rosemary essential oil supplementation, over a 200-day feeding period.
  • No significant differences in growth rates or flesh quality were observed among the different diet groups, but rosemary oil notably reduced malondialdehyde (MDA) levels, extending the fish's shelf life by up to 10 days.
  • The sensory quality of the fish fillets changed over time, with cooked fillet scores decreasing while fresh fillet scores increased, and the economic analysis suggested that using olive oil as a partial fish oil replacement is cost-effective.

Hemophilia is an X-linked congenital bleeding disorder for which factor replacement is life-saving but complicated by the sequelae of chronic hepatitis C virus (HCV) infection acquired decades ago. Although antiviral therapy clears HCV and reduces end-stage liver disease (ESLD), it may not reverse cirrhosis or prevent hepatocellular cancer (HCC). This was a retrospective cohort study of 121 men with hemophilia and HCV infection cared for at the Hemophilia Center of Western Pennsylvania to determine the incidence and predictors of ESLD and HCC.

Background: Almost half of severe hemophilia A (HA) cases are caused by an intron 22 inversion (Int22Inv) mutation, which truncates the 26-exon F8 messenger RNA (mRNA) after exon 22. Another F8 transcript, F8, is initiated from within F8-intron-22. F8 mRNA consists of a short exon spliced to exons 23 to 26 and is expressed in multiple human cell types.

  • - Podolian cattle are well-suited to tough areas in Southern Italy and are raised by letting them graze on natural pastures, which helps their health and the quality of the meat they produce.
  • - The study explored how the type of pasture (grassland or woodland) and the age when the bulls were slaughtered (14 or 18 months) affected their weight and the quality of the meat.
  • - Results showed that bulls grazing on grasslands were heavier and produced better meat quality at 18 months, and letting the meat age for 9 days improved tenderness without increasing harmful substances.

Gene therapy for severe hemophilia A uses an adeno-associated virus (AAV) vector and liver-specific promoters that depend on healthy hepatocyte function to achieve safe and long-lasting increases in factor VIII (FVIII) activity. Thus, hepatocyte health is an essential aspect of safe and successful gene therapy. Many people living with hemophilia A have current or past chronic hepatitis C virus infection, metabolic dysfunction-associated steatosis or steatohepatitis, or other conditions that may compromise the efficacy and safety of AAV-mediated gene therapy.

Article Synopsis
  • This study aimed to identify sources of acetylcholine (ACh) in white adipose tissue (WAT) and to investigate the activation of a non-neuronal cholinergic system in obese, inflamed WAT.
  • Diet-induced obesity was found to increase the expression of choline acetyltransferase (ChAT), mainly in macrophages within mouse epididymal WAT, suggesting these immune cells might produce ACh.
  • In human adipocytes, ACh reduced inflammatory markers and enhanced glucose uptake, indicating a potential therapeutic role for ACh in obesity-related inflammation.

Introduction: Gene therapy is now a reality for individuals with haemophilia, yet little is known regarding the quality-of-life impact of factor correction. As few data exist, and recognizing the analogy to liver transplantation (OLTX), we identified OLTX+ and OLTX- men in the ATHNdataset to compare post-OLTX factor VIII and IX on quality of life (QoL) by Haem-A-QoL and PROMIS-29.

Methods: OLTX- were matched to OLTX+ by age, race, and haemophilia type and severity.

Background: Prophylactic factor replacement therapy is recommended over on-demand treatment for preserving long-term joint health in hemophilia. Extended half-life products, including efmoroctocog alfa/eftrenonacog alfa (recombinant factor VIII [FVIII]/FIX Fc fusion proteins; herein rFVIIIFc/rFIXFc), have the potential to reduce treatment burden with less frequent administration and improve bleed prevention.

Objectives: We report post hoc data from patients with hemophilia A or B (HA/HB) who switched from prestudy on-demand FVIII/FIX to rFVIIIFc/rFIXFc prophylaxis at the start of A-LONG/B-LONG or start of/during ASPIRE/B-YOND phase 3 studies.
