This study focuses on enhancing the mechanical properties of thin, soft, free-standing films via a layer-by-layer (LBL) fabrication process called LBL-FP. Soft polymer nanocomposite (PNC) thin films, combining polydimethylsiloxane (PDMS) and single-walled carbon nanotubes (SWCNT) at ultra-low loadings using a unique bottom-up LBL-FP, are examined. Localized electrical breakdown (BD) measurements are performed on 2D muscovite mica flakes of ~ 2 to 15 nm thickness using Conduction Atomic Force Microscopy (CAFM). To obtain robust BD data by CAFM, the probed locations are spaced sufficiently far apart (> 1 µm) to avoid mutual interference and the maximum current is set to a low value (< 1 nA) to ensure severe damage does not occur to the sample. The analyses reveals that 2D muscovite mica has high electrical breakdown strength (12 MV/cm or more) and low leakage current, comparable to 2D hexagonal boron nitride (h-BN) of similar thickness.

We describe the synthesis of triazole-containing carboline derivatives and their utility as bromodomain and extra-terminal (BET) inhibitors. A convergent synthetic route permitted the detailed investigation of deuteration and fluorination strategies to reduce clearance while maintaining a favorable profile. This work led to the identification of a potent BET inhibitor, 2-{8-fluoro-3-[4-(H)methyl-1-methyl-1-1,2,3-triazol-5-yl]-5-[()-(oxan-4-yl)(phenyl)methyl]-5-pyrido[3,2-]indol-7-yl}propan-2-ol (), which demonstrated reduced clearance and an improved pharmacokinetic (PK) profile across preclinical species.

Resistive switching (RS) devices are emerging electronic components that could have applications in multiple types of integrated circuits, including electronic memories, true random number generators, radiofrequency switches, neuromorphic vision sensors, and artificial neural networks. Inhibition of the bromodomain and extra-terminal (BET) family of adaptor proteins is an attractive strategy for targeting transcriptional regulation of key oncogenes, such as c-MYC. Starting with the screening hit , a combination of structure-activity relationship and protein structure-guided drug design led to the discovery of a differently oriented carbazole with favorable binding to the tryptophan, proline, and phenylalanine (WPF) shelf conserved in the BET family.
