In this work we report the electrochemical, spectroscopical and spectro-electrochemical studies of a model complex [Cu(Bztpen)], (Bztpen = (N-benzyl-N,N',N'-tris(pyridin-2-ylmethyl)ethylenediamine) in order to propose a methodology to evaluate the interaction of potential metal based anticancer agents during electron transfer processes, with transport proteins such as Bovine Serum Albumin (BSA). It was possible to establish a reversible electron transfer [Cu(Bztpen)] +1e → [Cu(Bztpen)] and a weak interaction energy between BSA and [Cu(Bztpen)] and [Cu(Bztpen)] species, with no adsorption of protein over the electrode surface. Circular Dichroism (CD) Spectroelectrochemistry, not reported before, reveals no significant changes in BSA structure during the electron transfer [Cu(Bztpen)] + 1e → [Cu(Bztpen)].

For 156 2,2'-bipyridine derivatives reported in the academic literature, we calculated the redox potential, the pKa for the second deprotonation reaction, and the solubility in aqueous solutions. Using experimental data on a small subset of derivatives, we were able to calibrate our calculations.

View Article and Find Full Text PDFThe highly stable nitrosyl iron(II) mononuclear complex [Fe(bztpen)(NO)](PF6 )2 (bztpen=N-benzyl-N,N',N'-tris(2-pyridylmethyl)ethylenediamine) displays an S=1/2↔S=3/2 spin crossover (SCO) behavior (T1/2 =370 K, ΔH=12.48 kJ mol(-1) , ΔS=33 J K(-1) mol(-1) ) stemming from strong magnetic coupling between the NO radical (S=1/2) and thermally interconverted (S=0↔S=2) ferrous spin states. The crystal structure of this robust complex has been investigated in the temperature range 120-420 K affording a detailed picture of how the electronic distribution of the t2g -eg orbitals modulates the structure of the {FeNO}(7) bond, providing valuable magneto-structural and spectroscopic correlations and DFT analysis.

The [Fe(III)N5O] coordination core is constituted of the pentadentate ligand bztpen (N5) and a series of alkoxide anions (ethoxide, propoxide, n-butoxide, isobutoxide, and ethylene glycoxide). The methoxide derivative previously reported by us is also reinvestigated.

A complete study of the electronic density distribution in antineoplastic mixed chelate complexes of the type [Cu(N-N)(glycinate)H2 O]NO3 (N-N=2,2'-bipyridine (bpy) (1), 4,4'-dimethyl-bpy (2), 5,5'-dimethyl-2,2'-bipyridine (3), 1,10-phenanthroline (phen) (4), 4-methyl-phen (5); 5-methyl-phen (6); 4,7-dimethyl-phen (7), 5,6-dimethyl-phen (8), and 3,4,7,8-tetramethyl-phen (9)), a family known as Casiopeínas, was carried out with cyclic voltammetry, EPR, and computational methods. Crystal structures of 1⋅H2 O, 2⋅H2 O, 3⋅H2 O, 6⋅H2 O, and 8⋅H2 O show the variability in the geometries adopted by the copper compounds in the solid state. Experimental properties are described employing electronic descriptors obtained from computational methods.

Molecular dynamics were used to find the preferred site of binding between selected DNA chains and [Cu(2,2'-bipyridine)(acetylacetonate)(H(2)O)](+) (Cas). Full DFT optimizations of selected geometries extracted from simulations, followed by a topological analysis of electron density allowed us to define the specific interactions inside the recognition site.

A new synthetic pathway was reported to obtain N6 donor ligand 2,9-bis-(2',5'-diazahexanyl)-1,10-phenanthroline (L1) and its coordination compounds of essential divalent metal ions Mn, Fe, Co, Ni, Cu and Zn. Complete characterization of all compounds was done with the conventional techniques. Crystal structures of [NiL1](PF(6))(2) and [ZnL1](PF(6))(2)·H(2)O were also reported.
View Article and Find Full Text PDFCasiopeínas® are copper complexes with the general formula [Cu(N-N)(N-O)]NO(3) and [Cu(N-N)(O-O)]NO(3) where N-N denotes a substituted bipyridine or phenanthroline, N-O indicates α-aminoacidate or peptide and O-O represents acetylacetonate or salicylaldehyde. This family of compounds has been evaluated in vitro and in vivo showing cytotoxic, genotoxic, and antineoplastic activity. The action mechanism is still not completely elucidated, but the possibility exists that these compounds interact with DNA by intercalation due to the aromatic moiety.
View Article and Find Full Text PDFIn addition to a previous report, the reaction of tigogenin acetate with ICl in refluxing CHCl(3) produced the hitherto unknown 23R-iodotigogenin acetate, bearing an axial iodine atom at C-23 and its already reported 23S-epimer. The same treatment of sarsasapogenin acetate led to a single diasteromer characterized as 23S-iodosarsasapogenin acetate. A full characterization of the obtained compound including (1)H, (13)C NMR, MS and X-ray diffraction is provided.

The structural and solution characterization of novel Fe(II) compounds of the general formula [Fe(bztpen)X]PF6 and [Fe(bztpen)CH3CN](PF6)2 is presented, where bztpen is the pentadentate ligand N-benzyl-N,N',N'-tris(2-methylpyridyl)ethylenediamine and X- is a monodentate ligand. All complexes were characterized in solution and in the solid state, employing the usual techniques and single-crystal X-ray diffraction. The results obtained are discussed in terms of the existing information for some previously reported analogous compounds to arrive at a rationalization regarding the influence of a variation in the coordination environment of all compounds and to evaluate their relative stability.

View Article and Find Full Text PDFThe mononuclear diamagnetic compound {Fe(bztpen)[N(CN)2]}(PF6)CH3OH (1) (bztpen = N-benzyl-N,N',N'-tris(2-pyridylmethyl)ethylenediamine) has been synthesized and its crystal structure studied. Complex 1 can be considered to be the formal precursor of two new dinuclear, dicyanamide-bridged iron(II) complexes with the generic formula {[(Fe(bztpen)]2[mu-N(CN)2]}(PF6)3 x n H2O (n = 1 (2) or 0 (3)), which have been characterized in the solid state and in solution. In all three complexes, the iron atoms have a distorted [FeN6] octahedral coordination defined by a bztpen ligand and a terminal (1) or a bridging dicyanamide ligand (2 and 3).

Reaction of dichlorotris(triphenylphosphine) ruthenium(II) [RuCl(2)(PPh(3))(3)] with 1,8-bis(2-pyridyl)-3,6-dithiaoctane (pdto), a (N(2)S(2)) tetradentate donor, yields a new compound [Ru(pdto)(PPh(3))Cl]Cl (1), which has been fully characterized. (1)H and (31)P NMR studies of 1 in acetonitrile at several temperatures show the substitution of both coordinated chloride and triphenylphosphine with two molecules of acetonitrile, as confirmed by the isolation of the complex [Ru(pdto)(CH(3)CN)(2)]Cl(2) (2). Cyclic voltammetric and spectroelectrochemical techniques allowed us to determine the electrochemical behavior of compound 1.
