Volume reconstruction of large tissue specimens from serial physical sections using confocal microscopy and correction of cutting deformations by elastic registration.

A set of methods leading to volume reconstruction of biological specimens larger than the field of view of a confocal laser scanning microscope (CLSM) is presented. Large tissue specimens are cut into thin physical slices and volume data sets are captured from all studied physical slices by CLSM. Overlapping spatial tiles of the same physical slice are stitched in horizontal direction.

Rebuilding of rat intestinal mucous epithelium in prenatal ontogenesis.

Morphological differentiation of the intestinal epithelium in the laboratory rat occurs between the 16th and 21st day of prenatal development. The pseudostratified epithelium is rebuilt into simple epithelium of the future lining. A characteristic sign of this rebuilding is formation of primitive folds, villi and intraepithelial vacuoles corresponding in submicroscopic picture with a secondary luminization.

Apical ouabain-sensitive and ouabain-insensitive ATPases in rat colonic epithelium.

Active resorption of potassium ions in the colon is mediated by ouabain-sensitive and ouabain-insensitive H,K-ATPases localized in the apical membrane of colonic enterocytes (colonocytes). The present study was performed to investigate distribution patterns of apical ATPases using catalytic histochemistry and immunohistochemistry. Activity of the ATPases was localized both at the apical and basolateral regions of these cells.
