Circadian rhythmicity of gene expression is a conserved feature of cell physiology. This involves fine-tuning between transcriptional and post-transcriptional mechanisms and strongly depends on the metabolic state of the cell. Together these processes guarantee an adaptive plasticity of tissue-specific genetic programs.

Article Synopsis
  • Acoustically activated perfluoropropane droplets (PD) are used to enhance imaging of heart tissue, particularly in infarct zones, but their activation can be influenced by body temperature (BT).
  • The study analyzed the effects of BT during intravenous injection of PDs in rats, comparing temperatures above and below 36.5°C on myocardial contrast intensity (MCI) and microvascular retention.
  • Results showed that lower BT (<36.5°C) led to effective MCI enhancement in infarct zones, while higher BT inhibited activation and resulted in adverse effects on lung tissue.

Background: Mammalian testis is a highly complex and heterogeneous tissue. This complexity, which mostly derives from spermatogenic cells, is reflected at the transcriptional level, with the largest number of tissue-specific genes and long noncoding RNAs (lncRNAs) compared to other tissues, and one of the highest rates of alternative splicing. Although it is known that adequate alternative-splicing patterns and stage-specific isoforms are critical for successful spermatogenesis, so far only a very limited number of reports have addressed a detailed study of alternative splicing and isoforms along the different spermatogenic stages.

Background: Acoustically activatable perfluoropropane droplets (PD) can be formulated from commercially available microbubble preparations. Diagnostic transthoracic ultrasound frequencies have resulted in acoustic activation (AA) predominately within myocardial infarct zones (IZ).

Objective: We hypothesized that the AA area following acute coronary ischemia/reperfusion (I/R) would selectively enhance the developing scar zone, and target bioeffects specifically to this region.

Thromboangiitis obliterans (TAO), otherwise known as Buerger's disease, is a rare, non-atherosclerotic inflammatory vasculopathy that typically affects small and medium-sized arteries of the distal extremities. Smoking is believed to be integral to the pathogenesis, as TAO primarily affects young male smokers. The disease is characterized by extremity pain secondary to ischemia that may progress to ulceration, gangrene, and amputation.

  • Escalation of industrial activity has increased pollutants like heavy metals (e.g., copper), raising concerns about their bio-accumulative effects and the genetic factors influencing copper tolerance.
  • This study examined Drosophila melanogaster populations across Europe, revealing that latitude and urbanization significantly impacted copper tolerance, while known copper-related genes showed no differential expression between tolerant and sensitive strains.
  • Major physiological responses to copper toxicity were observed in the midgut, with gut acidity preservation linked to tolerance, and the study identified new genes and transposable elements that contribute to the copper stress response.

Background: Hydroxychloroquine is an effective and widely used treatment in multiple autoimmune connective tissue diseases that gained a lot of publicity in the coronavirus disease 2019 (COVID-19) pandemic. Our case reports are unique in that they explore the rare and sometimes overlooked effects of this drug on multiple organ systems, specifically the kidney, cardiac muscle, and skeletal muscle. We include key histologic features in images which aid in identifying and distinguishing hydroxychloroquine toxicity from mimickers.

As the coronavirus disease 2019 (COVID-19) pandemic evolves, much evidence implicates the heart as a critical target of injury in patients. The mechanism(s) of cardiac involvement has not been fully elucidated, although evidence of direct virus-mediated injury, thromboembolism with ischemic complications, and cytokine storm has been reported. We examined suggested mechanisms of COVID-19-associated heart failure in 21 COVID-19-positive decedents, obtained through standard autopsy procedure, compared to clinically matched controls and patients with various etiologies of viral myocarditis.

High quality reference genomes are crucial to understanding genome function, structure and evolution. The availability of reference genomes has allowed us to start inferring the role of genetic variation in biology, disease, and biodiversity conservation. However, analyses across organisms demonstrate that a single reference genome is not enough to capture the global genetic diversity present in populations.

COVID-19, the syndrome caused by the novel coronavirus SARS-CoV-2, has spread throughout the world, causing the death of at least three million people. For the over 81 million who have recovered, however, the long-term effects are only beginning to manifest. We performed a bilateral lung transplant on a 31-year-old male patient for chronic hypoxic respiratory failure, severe pulmonary hypertension and radiographically identified pulmonary fibrosis five months after an acute COVID-19 infection.

Article Synopsis
  • Trypanosomatids primarily manage gene expression after transcription, focusing on mRNA processing and translation control, unlike most eukaryotes, which mainly regulate synthesis via phosphorylation of eIF2.
  • In T. cruzi, the causative agent of Chagas' disease, we found that levels of eIF2α decrease in infective forms, indicating reduced protein synthesis and a rise in phosphorylation in proliferative stages before transforming into infective stages.
  • Mutations or overexpression of eIF2α led to significant changes in the parasite's proteins, resulting in less production of infective trypomastigotes, alongside alterations in surface protein composition that may affect infection capabilities.

Differences in social status are often mediated by agonistic encounters between competitors. Robust literature has examined social status-dependent brain gene expression profiles across vertebrates, yet social status and reproductive state are often confounded. It has therefore been challenging to identify the neuromolecular mechanisms underlying social status independent of reproductive state.

It is generally accepted that the presence of ORFs in the 5' untranslated region of eukaryotic transcripts modulates the production of proteins by controlling the translation initiation rate of the main CDS. In trypanosomatid parasites, which almost exclusively depend on post-transcriptional mechanisms to regulate gene expression, translation has been identified as a key step. However, the mechanisms of control of translation are not fully understood.
