Dilated cardiomyopathy (DCM) is a highly prevalent and genetically heterogeneous condition that results in decreased contractility and impaired cardiac function. The FK506-binding protein FKBP12 has been implicated in regulating the ryanodine receptor in skeletal muscle, but its role in cardiac muscle remains unclear. To define the effect of FKBP12 in cardiac function, we generated conditional mouse models of FKBP12 deficiency.

Here we show that striated muscle preferentially expressed protein kinase α (Spegα) maintains cardiac function in hearts with Spegβ deficiency. Speg is required for stability of excitation-contraction coupling (ECC) complexes and interacts with esterase D (Esd), Cardiomyopathy-Associated Protein 5 (Cmya5), and Fibronectin Type III and SPRY Domain Containing 2 (Fsd2) in cardiac and skeletal muscle. Mice with a sequence encoding a V5/HA tag inserted into the first exon of the Speg gene (HA-Speg mice) display a >90% decrease in Spegβ but Spegα is expressed at ~50% of normal levels.

Mutations in the skeletal muscle Ca release channel, the type 1 ryanodine receptor (RYR1), cause malignant hyperthermia susceptibility (MHS) and a life-threatening sensitivity to heat, which is most severe in children. Mice with an MHS-associated mutation in Ryr1 (Y524S, YS) display lethal muscle contractures in response to heat. Here we show that the heat response in the YS mice is exacerbated by brown fat adaptive thermogenesis.

Biosourced materials are gaining interest industrially, but there are still limitations on the library of available materials suitable for advanced manufacturing, especially using photopolymerization-based processing techniques. Terpenes, such as myrcene, are naturally produced materials possessing structural features, specifically alkenes, that avail themselves for such techniques. Free-radical and anionic polymerization techniques were used to explore molecular architecture, such as branching, as well as molecular weight and dispersity on physical properties prior to the production of 3D printing photopolymer resins.

A mechanically strong composite hydrogel was produced based on an interpenetrating network (IPN) between gelatin and silk fibroin. When two layers of the IPN were created, the resulting hydrogel exhibited much improved mechanical properties. This hydrogel is biodegradable and non-cytotoxic and allows for cell adhesion and proliferation on the surface.

A review of the literature published in 2016 on topics relating to water reclamation and reuse is summarized. The review is divided into the following sections: 1) General: monitoring and assessment, social awareness and acceptance, and energy and resources recovery; 2) Treatment technology: integrated technologies, membrane treatment, reactors, WSPs, disinfection, and others; 3) Planning and Management: modeling, LCA, GHG emission, and case studies. Much of the water treatment research focused on the integrated technologies and membrane treatment.

The electrogenic sodium/calcium exchanger (NCX) mediates bidirectional calcium transport controlled by the transmembrane sodium gradient. NCX inactivation occurs in the absence of phosphatidylinositol 4,5-bisphosphate and is facilitated by palmitoylation of a single cysteine at position 739 within the large intracellular loop of NCX. The aim of this investigation was to identify the structural determinants of NCX1 palmitoylation.
