To characterize the genomic features of a community-acquired Acinetobacter baumannii strain, co-carrying tet(X6) and bla genes, but was susceptible to tigecycline and carbapenems. The tet(X6) and bla genes were found on a 149,518 bp non-conjugative plasmid. The bla gene was silent, due to the presence of an intact ISAba3-like element upstream, which rendered the strain susceptible to carbapenems.

The development of prophylactic cancer vaccines typically involves the selection of combinations of tumour-associated antigens, tumour-specific antigens and neoantigens. Here we show that membranes from induced pluripotent stem cells can serve as a tumour-antigen pool, and that a nanoparticle vaccine consisting of self-assembled commercial adjuvants wrapped by such membranes robustly stimulated innate immunity, evaded antigen-specific tolerance and activated B-cell and T-cell responses, which were mediated by epitopes from the abundant number of antigens shared between the membranes of tumour cells and pluripotent stem cells. In mice, the vaccine elicited systemic antitumour memory T-cell and B-cell responses as well as tumour-specific immune responses after a tumour challenge, and inhibited the progression of melanoma, colon cancer, breast cancer and post-operative lung metastases.

The recent worldwide outbreaks of mpox prioritize the development of a safe and effective mRNA vaccine. The contemporary mpox virus (MPXV) exhibits changing virological and epidemiological features, notably affecting populations already vulnerable to human immunodeficiency virus (HIV). Herein, we profile the immunogenicity of AR-MPXV5, a penta-component mRNA vaccine targeting five specific proteins (M1R, E8L, A29L, A35R, and B6R) from the representative contemporary MPXV clade II strain, in both naive and simian immunodeficiency virus (SIV)-infected nonhuman primates.

Background: The effect of embolization of the middle meningeal artery in patients with subacute or chronic subdural hematoma is uncertain.

Methods: We performed a multicenter, open-label, randomized trial in China, involving patients with symptomatic nonacute subdural hematoma with mass effect. Patients were assigned to undergo burr-hole drainage or receive nonsurgical treatment at the surgeon's discretion, and patients in each group were then randomly assigned, in a 1:1 ratio, to undergo middle meningeal artery embolization with liquid embolic material or to receive usual care.

Article Synopsis
  • A study was conducted to find out the best systolic blood pressure targets for patients over 50 with type 2 diabetes and high cardiovascular risk, comparing intensive treatment (target <120 mm Hg) to standard treatment (target <140 mm Hg) over 5 years.
  • Out of 12,821 patients, the intensive treatment group had a lower average systolic blood pressure after one year (121.6 mm Hg) compared to the standard group (133.2 mm Hg), resulting in significantly fewer major cardiovascular events.
  • Despite the lower overall events in the intensive group, there were more cases of symptomatic hypotension and hyperkalemia, but the rates of serious adverse events were similar in both groups.

The storage of hydrogen is key to its applications. Developing adsorbent materials with high volumetric and gravimetric storage capacities, both of which are essential for the efficient use of hydrogen as a fuel, is challenging. Here we report a controlled catenation strategy in hydrogen-bonded organic frameworks (RP-H100 and RP-H101) that depends on multiple hydrogen bonds to guide catenation in a point-contact manner, resulting in high volumetric and gravimetric surface areas, robustness and ideal pore diameters (~1.

The precise control of and configurations of the carbon-carbon double bond in alkene synthesis has long been a fundamental challenge in synthetic chemistry, even more pronounced when simultaneously striving to achieve enantioselectivity [(,), (,), (,), (,)]. Moreover, enantiopure non-natural α-amino acids are highly sought after in organic and medicinal chemistry. In this study, we report a ligand-controlled stereodivergent synthesis of non-natural α-quaternary amino acids bearing trisubstituted alkene moieties in high yields with excellent enantioselectivity and / selectivities.

  • CAR T therapy is effective for blood cancers but struggles with solid tumors like glioblastoma due to the immunosuppressive tumor microenvironment (TME).
  • The transforming growth factor β (TGF-β) is a key factor in promoting this suppressive environment, hindering immune responses against GBM.
  • Researchers are developing a new CAR T construct (CART-EGFR-IL13Rα2-dnTGFβRII) that combines a modified TGF-β receptor with existing therapy to improve T cell function and target GBM more effectively, showing promise in both lab studies and mouse models.

Here, we report a strategy enabling triple switchable chemo-, regio-, and stereodivergence in newly developed palladium-catalyzed cycloadditions of allenes. An asymmetric pseudo-stereodivergent cycloaddition of allenes bearing a primary leaving group at the α-position, where a dynamic kinetic asymmetric hydroalkoxylation of racemic unactivated allenes was the enantio-determining step, is realized, providing four stereoisomers [(Z,R), (Z,S), (E,S), and (E,R)] containing a di-substituted alkene scaffold and a stereogenic center. By tuning reaction conditions, a mechanistically distinctive cycloaddition is uncovered selectively with the same set of substrates.
