Effective antitumor T cell activity relies on the expression and MHC presentation of tumor neoantigens. Tumor cells can evade T cell detection by silencing the transcription of antigens or by altering MHC machinery, resulting in inadequate neoantigen-specific T cell activation. We identified the DNA-protein kinase inhibitor (DNA-PKi) NU7441 as a promising immunomodulator that reduced immunosuppressive proteins, while increasing MHC-I expression in a panel of human melanoma cell lines.

Inclisiran is a novel small interfering RNA targeting proprotein convertase subtilisin/kexin type 9 (PCSK9) that was approved by the US FDA in December 2021. After two doses three months apart, it is administered biannually as a subcutaneous injection and has been shown to lower LDL-C by ∼50 % in clinical trials. Here, we present real-world data on the prescription and administration of inclisiran at the University of Pennsylvania Health Systems.

Article Synopsis
  • Constrictive pericarditis is a condition caused by chronic inflammation of the pericardium, with tuberculosis being a common cause outside the U.S. and a serious complication of pericardial disease.
  • A case study as illustrated by a 31-year-old man who experienced symptoms like chest pain and dyspnea, leading to diagnoses through various tests, including an echocardiogram and right heart catheterization.
  • The patient's treatment involved surgery to remove thickened pericardium and a 6-month antibiotic therapy regimen, underscoring the need for a multidisciplinary approach in managing this condition, especially in cases tied to tuberculosis.

Background: Residential aged-care facilities (RACFs, also called long-term care facilities, aged care homes, or nursing homes) have elevated risks of respiratory infection outbreaks and associated disease burden. During the COVID-19 pandemic, social isolation policies were commonly used in these facilities to prevent and mitigate outbreaks. We refer specifically to general isolation policies that were intended to reduce contact between residents, without regard to confirmed infection status.

Article Synopsis
  • Vamorolone, a new steroid, showed similar benefits to prednisone for muscle function in kids with Duchenne muscular dystrophy (DMD) while promoting better growth and bone health in trials.
  • The study aimed to compare how often adrenal suppression (AS) occurred with vamorolone versus prednisone in young patients and to evaluate cortisol levels using a specific immunoassay.
  • Results indicated that AS was common in both treatments, particularly with higher doses of vamorolone, and suggested that a lower cortisol level threshold may be needed for accurate assessment, recommending hydrocortisone for stress dosing.
