Background: Conventional photodynamic therapy (cPDT) is an effective treatment option for field cancerization and multiple actinic keratoses (AK). The main side effect of cPDT is pain during illumination which in severe cases might necessitate early termination of treatment. Modification of treatment parameters such as light dose and fluence rate is a promising approach to mitigate PDT-associated pain.

Article Synopsis
  • Treatment switching in clinical trials complicates causal inference, making intention-to-treat (ITT) analyses less reliable for determining the true treatment effect.
  • The ALTA-1L trial on brigatinib versus crizotinib highlighted this issue, showing more promising results for brigatinib when removed from treatment switching scenarios, via methods like Inverse Probability of Censoring Weights (IPCW).
  • Re-analysis using IPCW and g-methods suggested that the treatment effect of brigatinib on overall survival may be significantly better than previously estimated, indicating that standard ITT analysis might underestimate treatment benefits.

Article Synopsis
  • The study evaluates the real-world effectiveness of two oral antivirals, nirmatrelvir-ritonavir and molnupiravir, against the Omicron variant of SARS-CoV-2 in non-hospitalized adults.
  • Data were obtained from a retrospective cohort study involving over 113,000 confirmed cases of Covid-19, comparing treated patients with untreated controls to assess risks of hospitalization and death within 28 days.
  • Results showed that nirmatrelvir-ritonavir significantly reduced hospitalization and death rates, especially in individuals aged 60 and older; however, molnupiravir did not show a significant difference in these outcomes compared to untreated controls.

Measuring disease progression in clinical trials for testing novel treatments for multifaceted diseases as progressive supranuclear palsy (PSP), remains challenging. In this study we assess a range of statistical approaches to compare outcomes as measured by the items of the progressive supranuclear palsy rating scale (PSPRS). We consider several statistical approaches, including sum scores, a modified PSPRS rating scale that had been recommended by FDA in a pre-IND meeting, multivariate tests, and analysis approaches based on multiple comparisons of the individual items.

Background: Corroborating evidence for the use of hypothermic oxygenated machine perfusion (HOPE) prior to orthotopic liver transplantation (OLT) suggests a beneficial effect in regard to biliary complications. Here, the authors aim to evaluate whether perfusion via portal vein alone (sHOPE) or via additional perfusion of the hepatic artery (dHOPE) have diverging impact on outcomes after OLT when compared to the use of static cold storage (SCS).

Methods: Consecutive patients undergoing OLT at the Medical University of Vienna (2018-2023) were retrospectively analyzed.
