The fermentation of desmethyl-asterriquinone B-1, a diabetes target, by a Pseudomonasarias species was conducted at the 600-l scale using a revised complex medium containing yeast extract and soy hydrolysate. Oat flour and tomato paste were removed from this medium due to difficulties in sterilization. An initial cerelose charge of 40 g/l improved titer and reduced product degradation in the broth at cultivation conditions.

Article Synopsis
  • A new method was developed to produce toluene cis glycol using a specific strain of Rhodococcus (MA 7249) lacking certain enzymes.
  • The produced toluene cis glycol exhibited optical rotations that confirmed its absolute configuration as (1S,2R).
  • When grown in a lab with toluene vapors, MA 7249 achieved a concentration of 18 g/l of toluene cis glycol in about 110 hours and also produced similar dihydrodiols from other compounds.

Article Synopsis
  • A rapid microbial screening method was developed using a 24-well plate format, automated liquid handling, and HPLC to evaluate bioconversion of 6-bromo-beta-tetralone to 6-bromo-beta-tetralol.
  • The method utilized pre-selected yeast and rhodococci strains, ensuring a high "hit rate" for bioreduction activity.
  • Results showed that 78% of the 240 strains screened successfully bioconverted the target compound, demonstrating efficiency over traditional larger volume screening methods.

Article Synopsis
  • Cis (1S,2R) indanediol is a crucial precursor in creating (-)-cis (1S,2R)-1-aminoindan-2-ol, which is essential for the HIV protease inhibitor Crixivan.
  • A study identified the yeast strain Trichosporon cutaneum MY 1506 as an effective biocatalyst for converting 1,2-indanedione into cis (1S,2R) indanediol, achieving an impressive yield of 99.1% and over 99% enantiomeric excess in initial tests.
  • When the process was scaled up to a 23-liter bioreactor, the strain produced 8.4 grams of pure
