Genome-wide association studies (GWAS) have identified more than a hundred single nucleotide variants (SNV) associated with the risk of gastroesophageal cancer (GEC). The majority of the identified SNVs map to noncoding regions of the genome. Uncovering the causal SNVs and genes they modulate could help improve GEC prevention and treatment.

Septins are a family of cytoskeletal proteins that regulate several important aspects of neuronal development. Septin 7 (Sept7) is enriched at the base of dendritic spines in excitatory neurons and mediates both spine formation and spine and synapse maturation. Phosphorylation at a conserved C-terminal tail residue of Sept7 mediates its translocation into the dendritic spine head to allow spine and synapse maturation.

Article Synopsis
  • The study aimed to compare how pharmacists and internal medicine providers prescribe statins to diabetes patients aged 40 and older, analyzing healthcare records from 2017.
  • Results showed that patients managed by pharmacists were prescribed appropriate statin therapy more often than those managed by internal medicine providers, especially for high-intensity statins and certain demographic groups such as younger females.
  • Despite the findings, overall statin prescribing for diabetes patients was still low, indicating a need for improvement in optimizing therapy for these patients.

Pharmacological strategies that boost intracellular NAD are highly coveted for their therapeutic potential. One approach is activation of nicotinamide phosphoribosyltransferase (NAMPT) to increase production of nicotinamide mononucleotide (NMN), the predominant NAD precursor in mammalian cells. A high-throughput screen for NAMPT activators and hit-to-lead campaign yielded SBI-797812, a compound that is structurally similar to active-site directed NAMPT inhibitors and blocks binding of these inhibitors to NAMPT.
