The Retriever complex recycles a wide range of transmembrane proteins from endosomes to the plasma membrane. The cargo adapter protein SNX17 has been implicated in recruiting the Retriever complex to endosomal membranes, yet the details of this interaction have remained elusive. Through biophysical and structural model-guided mutagenesis studies with recombinant proteins and liposomes, we have gained a deeper understanding of this process.

This is accompanied by activation of a pro-inflammatory gene expression programme - the senescence-associated secretory phenotype (SASP) - driven by transcription factors such as NF-κB. The relationship between heterochromatin re-organisation and the SASP has been unclear.

Because ERK2 can also bind to DNA to modify gene expression, we investigated whether it more directly regulates transcription. We identified ERK2 binding sites in the promoter and detected ERK2 at the promoter in various serum-stimulated cell types.

Despite growing interest in targeted elimination of senescent cells, only few senolytics are known due to the lack of well-characterised molecular targets. Here, we report the discovery of three senolytics using cost-effective machine learning algorithms trained solely on published data.

In this paper, two different three-dimensional (3D) Fe/SiC catalysts with a square cell geometry have been manufactured by Robocasting: monoliths ( = 14 and = 15 mm) and meshes ( = 24 and = 2 mm) and studied in the catalytic phenol oxidation by hydrogen peroxide (HO) for the sustainable production of dihydroxybenzenes (DHBZ). The fluid dynamics, catalytic performance, reaction rates, external mass transport limitation, and catalyst stability have been compared in three different reactors, monolithic fixed-bed reactor, multimesh fixed-bed reactor, and monolithic stirrer reactor, at selected operating conditions.

Here we show that LPS-mediated activation of caspase-4 also induces a stress response promoting cellular senescence, which is dependent on the caspase-4 substrate gasdermin-D and the tumor suppressor p53. Furthermore, we found that the caspase-4 noncanonical inflammasome is induced and assembled in response to oncogenic RAS signaling during oncogene-induced senescence (OIS).

MNT dimerizes with another transcriptional regulator, MYC-associated factor X (MAX), and down-regulates genes by binding to E-boxes. MAX also dimerizes with MYC, an oncogenic bHLH transcription factor.

Here, we show that, during oncogene-induced senescence (OIS), the Toll-like receptor 2 (TLR2) and its partner TLR10 are key mediators of senescence in vitro and in murine models. TLR2 promotes cell cycle arrest by regulating the tumor suppressors p53-p21, p16, and p15 and regulates the SASP through the induction of the acute-phase serum amyloids A1 and A2 (A-SAAs) that, in turn, function as the damage-associated molecular patterns (DAMPs) signaling through TLR2 in OIS.

Senescence can be organismally beneficial as it can prevent the propagation of damaged or mutated clones and stimulate their clearance by immune cells. However, it has recently become clear that senescence also contributes to the pathophysiology of aging through the accumulation of damaged cells within tissues.

The ubiquitination reaction is catalyzed by a multitude of ligases, whereas the removal of ubiquitin chains is mediated by two deubiquitinating enzymes (DUBs), USP7 (HAUSP) and USP10. Here, we show that PHD3 hydroxylates p53 at proline 359, a residue that is in the p53-DUB binding domain.

In this study we used vacuum impregnation (VI) methods to incorporate pectin and calcium chloride into whole red raspberries to improve their firmness. Specifically, we impregnated low methoxyl pectin (LMP) at 10 g of pectin kg of solution and calcium chloride (CaCl ·2H O) at 30 g calcium kg of pectin, and on the other side pectin methylesterase (PME) at 10 g of enzyme kg of solution, and (CaCl ·2H O) at 10 g of calcium kg of solution, into whole red raspberries.

In the last few years, several works dealing with Fenton oxidation of ionic liquids (ILs) have proved the capability of this technology for their degradation, achieving complete ILs removal and non-toxic effluents. Nevertheless, very little is known about the kinetics of this process, crucial for its potential application. In this work, the effect of several operating conditions, including reaction temperature (50-90 °C), catalyst load (10-50 mg L Fe), initial IL concentration (100-2000 mg L), and hydrogen peroxide dose (10-200% of the stoichiometric amount for the complete IL mineralization) on 1-butyl-3-methylimidazolium chloride ([Cmim]Cl) oxidation has been investigated.

Despite the increasing popularity of 3D printing, also known as additive manufacturing (AM), the technique has not developed beyond the realm of rapid prototyping. This confinement of the field can be attributed to the inherent flaws of layer-by-layer printing and, in particular, anisotropic mechanical properties that depend on print direction, visible by the staircasing surface finish effect. Continuous liquid interface production (CLIP) is an alternative approach to AM that capitalizes on the fundamental principle of oxygen-inhibited photopolymerization to generate a continual liquid interface of uncured resin between the growing part and the exposure window.

Unravelling the changes that occur in the chromatin structure during these processes has been a topic of interest for many years. This work deals with the evolution of intermediates and ecotoxicity upon Fenton's oxidation of phenol in aqueous solution. The EC50 values of the intermediates identified in the oxidation pathway of phenol have been measured. Some of these compounds, mainly hydroquinone and p-benzoquinone, showed toxicity levels much higher than phenol itself.
