Mulberry crinkle leaf virus (MCLV) is a representative species of the genus Mulcrilevirus in the family Geminiviridae. Here, we identified an additional V6 ORF which embedded within the V4 ORF in the MCLV virion-sense strand. The expression of V6 was confirmed by analyzing the promoter activity of V6 ORF upstream sequences and quantifying the viral DNA accumulation in V6-mutant MCLV-infected tomato plants.

The primer pairs were designed based on the conserved sequence of 25 HSVd variants deposited in the GenBank database. Four out of a total of 53 samples were HSVd-positive, confirming that HSVd is present in mulberry plants in China.

The sequence consisted of 2952 nt, containing four open reading frames (ORFs) in the viral-sense strand and two ORFs in the complementary-sense strand. The size of the genome and the conserved origin of replication are similar to those of members of the family Geminiviridae, but the genomic organization, number of ORFs, and especially five contiguous GAAAAA repeats positioned upstream of ORF1 distinguish it from other geminiviruses.
