l-Tryptophan decarboxylase (TDC) and l-3,4-dihydroxyphenylalanine decarboxylase (DDC) catalyze the decarboxylation of l-tryptophan, 5-hydroxytryptophan, and l-3,4-dihydroxyphenylalanine. In this study, we analyzed the amino acid compositions of the substrate-binding pockets of TDC from (TDC) and DDC from (DDC), explored the specificity of key amino acids within these pockets, and elucidated mechanisms influencing substrate selectivity and catalytic activity in both enzymes, using whole-cell catalysis to screen mutants and determine enzymatic kinetic parameters. The results demonstrated that residues Ala-103 and Val-122 in TDC, along with their corresponding sites Thr-82 and Ile-101 in DDC, significantly influence substrate selectivity and catalytic efficiency.

A rapid method for fatty acids (FAs) comparative profiling based on carboxyl-specific stable isotope labeling (SIL) and direct infusion electrospray ionization-ion mobility-mass spectrometry (ESI-IM-MS) is established. The design of the method takes advantage of the three-dimensional characteristics of IM-MS including drift time, m/z and ion intensity, for comparison of d0-/d6-2,4-dimethoxy-6-piperazin-1-yl pyrimidine (DMPP)-labeled FAs. In particular, without chromatographic separation, the method allowed direct FAs profiling in complex samples due to the advantageous priority of DMPP in signal enhancement as well as the extra resolution that IM-MS offered.

In this study, we confirmed the autophagy induced by porcine reproductive and respiratory syndrome virus (PRRSV) in permissive cells and investigated the role of autophagy in the replication of PRRSV. We first demonstrated that PRRSV infection significantly results in the increased double-membrane vesicles, the accumulation of LC3 fluorescence puncta, and the raised ratio of LC3-II/β-actin, in MARC-145 cells. Then we discovered that induction of autophagy by rapamycin significantly enhances the viral titers of PRRSV, while inhibition of autophagy by 3-MA and silencing of LC3 gene by siRNA reduces the yield of PRRSV.

A simple, sensitive and selective fluorimetric determination for trace nitrites was developed using an unsymmetrical rhodamine with a high fluorescence quantum yield and large Stokes shift. The method is based on the reaction of the unsymmetrical rhodamine with nitrite in an acidic medium to form a nitroso product, which has much lower fluorescence because the electron-withdrawing effect of the nitroso group influences the system of p-pi conjugation between N atom and benzene ring. Under optimum conditions, the fluorescence quenching intensity is linear over a nitrite concentration range of 1.
