The molecular mechanism underlying the role of hippocampal hilar interneuron degeneration in temporal lobe epilepsy (TLE) remains unclear. Especially, very few studies have focused on the role of neuronal nitric oxide synthase (nNOS, encoded by Nos1) containing hilar interneurons in TLE. In the present study, Nos1 conditional knockout mice were constructed, and we found that selective deletion of Nos1 in hilar interneurons rather than dentate granular cells (DGCs) triggered epileptogenesis.

Astrocytes, the predominant glial cell type in the mammalian brain, influence a wide variety of brain parameters including neuronal energy metabolism. Exciting recent studies have shown that obesity and diabetes can impact on astrocyte function. We review evidence that dysregulation of astrocytic lipid metabolism and glucose sensing contributes to dysregulation of whole-body energy balance, thermoregulation, and insulin sensitivity.

Glioblastoma, the most lethal primary brain tumor, harbors glioma stem cells (GSCs) that not only initiate and maintain malignant phenotypes but also enhance therapeutic resistance. Although frequently mutated in glioblastomas, the function and regulation of PTEN in PTEN-intact GSCs are unknown. Here, we found that PTEN directly interacted with MMS19 and competitively disrupted MMS19-based cytosolic iron-sulfur (Fe-S) cluster assembly (CIA) machinery in differentiated glioma cells.

Article Synopsis
  • Glucocorticoids (GCs) are essential hormones that help with cognitive functions, but both too much and too little can harm memory and mood, potentially linked to their U-shaped effect on the brain.
  • In experiments with mice, varying levels of GCs influenced the growth of neural stem cells (NSCs) in a U-shaped manner, promoting growth at normal levels but inhibiting it at higher stress levels.
  • Additionally, interventions like blocking glucocorticoid synthesis or surgically removing adrenal glands affected NSC development, and boosting a particular protein (telomerase reverse transcriptase) can help counteract cognitive deficits caused by stress or low GC levels.

Temozolomide (TMZ) is a standard treatment for glioblastoma (GBM) patients. However, TMZ has moderate therapeutic effects due to chemoresistance of GBM cells through less clarified mechanisms. Here, we demonstrate that TMZ-derived 5-aminoimidazole-4-carboxamide (AICA) is converted to AICA ribosyl-5-phosphate (AICAR) in GBM cells.

The effects of initial soil moisture on colloid-associated transport are still poorly understood given the well-recognized significance of colloid-facilitated transport of strongly-sorbing contaminants. In this study, Cd leaching was sequentially conducted in an intact soil column under three initial moisture conditions (near saturation, field capacity and dryness). Soil colloids were always the dominant carriers for Cd.

  • In response to stress, the HPA axis activates and releases glucocorticoids, which can negatively affect the body's stress response, especially in the hippocampus.
  • Maternal stress during pregnancy disrupts the normal feedback function of the HPA axis in offspring, leading to potential issues later in adulthood.
  • A study found that knocking out the TERT gene leads to an overactive HPA axis due to changes in the brain's hippocampus, rather than a lack of glucocorticoid receptors, suggesting new ways to understand stress impacts on the body.

Major depressive disorder (MDD) is one of the most common mental disorders. We designed a fast-onset antidepressant that works by disrupting the interaction between the serotonin transporter (SERT) and neuronal nitric oxide synthase (nNOS) in the dorsal raphe nucleus (DRN). Chronic unpredictable mild stress (CMS) selectively increased the SERT-nNOS complex in the DRN in mice.

Panic disorder (PD) causes serious functional damage and disability and accelerates the process of individual aging. The pathological basis of PD is the same as that of age-related diseases, which is proposed as a new viewpoint in recent years. Memory decline and social functional impairment are common manifestations of accelerated aging in PD.

Major depressive disorder (MDD) is one of the foremost causes of disability and premature death worldwide. Although the available antidepressants are effective and well tolerated, they also have many limitations. Therapeutic advances in developing a new drug's ultimate relation between MDD and chronobiology, which targets the circadian rhythm, led to a renewed focus on psychiatric disorders.

Article Synopsis
  • The exact cause of post-traumatic stress disorder (PTSD) is still not fully understood, but abnormal serotonin levels are thought to play a significant role.
  • Research indicates that nitric oxide (NO) impacts serotonin levels in the brain, although the effects are inconsistent and not well understood.
  • This study suggests that high levels of nitric oxide due to the activity of nNOS in the dorsal raphe nucleus suppress serotonin neuron activity, which may lead to PTSD-like symptoms such as increased anxiety and heightened fear responses.

Sucrose preference test (SPT) is a most frequently applied method for measuring anhedonia, a core symptom of depression, in rodents. However, the method of SPT still remains problematic mainly due to the primitive, irregular, and inaccurate various types of home-made equipment in laboratories, causing imprecise, inconsistent, and variable results. To overcome this issue, we devised a novel method for automatic detection of anhedonia in mice using an electronic apparatus with its program for automated detecting the behavior of drinking of mice instead of manual weighing the water bottles.

The gut microbiota is composed of a large number of microbes, usually regarded as commensal bacteria. It has become gradually clear that gastrointestinal microbiota affects gut pathophysiology and the central nervous system (CNS) function by modulating the signaling pathways of the microbiota-gut-brain (MGB) axis. This bidirectional MGB axis communication primarily acts through neuroendocrine, neuroimmune, and autonomic nervous systems (ANS) mechanisms.

The cerebrovascular system is not only inert bystandard that support the metabolic demands of the brain but also elicit the barrier functions against risk factors mediated neurovascular injury. The onsets of cerebrovascular inflammation are considered as stimuli that can provoke the host defense system and trigger the development of neurological disorders. Homeostasis of the brain function is regulated by the movement of endothelial, glial, and neuronal cells within the neurovascular unit (NVU), which acts as a "platform" for the coordinated action of anti- and pro-inflammatory mechanisms.
