Effect of deleterious nsSNP on the HER2 receptor based on stability and binding affinity with herceptin: a computational approach.

In this study, we identified the most deleterious non-synonymous SNP of ERBB2 (HER2) receptors by its stability and investigated its binding affinity with herceptin. Out of 135 SNPs, 10 are nsSNPs in the coding region, in which one of the nsSNP (SNPid rs4252633) is commonly found to be damaged by I-Mutant 2.0, SIFT and PolyPhen servers.

Studies on flexibility and binding affinity of Asp25 of HIV-1 protease mutants.

We have investigated and highlighted the behavior of binding residue, Asp25 by computational analysis, which play an important role in understanding docking process with drug molecule, Ritonavir (Norvir) and the flexibility nature of the Human Immunodeficiency Virus-1 (HIV-1) protease enzyme. It is well known that Ritonavir is a potent and a selective HIV-1 protease inhibitor. Molecular dockings were performed in order to gain insights regarding the binding mode of this inhibitor.

Identification and structural comparison of deleterious mutations in nsSNPs of ABL1 gene in chronic myeloid leukemia: a bio-informatics study.

Single nucleotide polymorphism (SNP) serve as frequent genetic markers along the chromosome. They can, however, have important consequences for individual susceptibility to disease and reactions to medical treatment. Also, genetics of the human phenotype variation could be understood by knowing the functions of these SNPs.

The pathophysiology of large capacity bladder.

Purpose: We describe the pathophysiology, differential diagnosis and urodynamic findings in patients with a large capacity bladder.

Materials And Methods: This was a retrospective, observational study of 100 consecutive patients with voiding dysfunction and a cystometric bladder capacity of greater than 700 ml. Clinical data, cystometric bladder capacity and other urodynamic findings were evaluated.

Nanostructured La(Sr)CrO3 through gel combustion: sintering and electrical behavior.

Nanostructured La(Sr)CrO3 (LSC) powders was prepared through glycine-nitrate gel combustion process. It was shown for the first time that the use of relatively inexpensive CrO3 as a starting material for chromium has a potential for the bulk preparation of sinter-active LSC powder. As-prepared powder when calcined at 700 degrees C resulted in LSC along with a small amount of SrCrO4 as a secondary phase.

Blood flow to the corpus cavernosum penis in the bull and goat buck during penile quiescence.

Bulls and bucks were used to study the blood flow into and out of the corpus cavernosum penis (CCP) during the non-erect state of the penis. When contrast medium was injected into the dorsal artery of the penis in a bull cadaver and into surgically implanted catheters of anesthetized bucks, it flowed into the cavernous spaces of the CCP via the penetrating arteries. When contrast medium was injected directly into the CCP of the bull and buck there was no evidence of vascular exits along the body of the penis.

Synthesis of nanocrystalline La(Ca)CrO3 through a novel gel combustion process and its characterization.

La(Ca)CrO3 (LCC) powders have been prepared through a novel gel combustion route using metal nitrates as oxidizers and ethylene diamine tetraacetic acid (EDTA) as a fuel. The process involves the combustion of a viscous gel obtained through mixed metal nitrate-EDTA solution. The effect of amount of EDTA used in the process on the nature of combustion reaction, phase formation, and densification has been studied.

Nanocrystalline ceria powders through citrate-nitrate combustion.

Nanocrystalline ceria powders have been synthesized by combustion technique using citric acid as a fuel and nitrate as an oxidizer. The auto-ignition of the gels containing cerium nitrate and citric acid resulted in ceria powders. A theory based on adiabatic flame temperature for different citric acid-to-cerium nitrate molar ratios has been proposed to explain the nature of combustion reaction and its correlation with the powder characteristics.

Signet ring cell carcinoma of ampulla of Vater in a young adult.

Signet ring cell carcinoma of the ampulla of Vater is extremely rare. The 7 cases reported earlier have been in older patients. We report a 32-year-old lady with this condition, who also had metastases in the bone marrow, vertebrae, lungs and liver.

Linear mRNA amplification from as little as 5 ng total RNA for global gene expression analysis.

Gene expression analysis has become an invaluable tool for understanding gene function and regulation. However, global expression analysis requires large RNA quantities or RNA preamplification. We describe an isothermal messenger RNA (mRNA) amplification method, Ribo-SPIA, which generates micrograms of labeled cDNA from 5 ng of total RNA in 1 day for analysis on arrays or by PCR quantification.

Stone clustering of patients with cystine urinary stone formation.

Objectives: To explore the history of symptomatic cystine stone formation. Cystinuria is a genetic defect that may result in the formation of recurrent cystine calculi.

Methods: Thirty-four patients with cystinuria were retrospectively evaluated for treatment patterns, medical prophylaxis, and renal outcome.

Update of staging and risk assessment for prostate cancer patients.

Purpose Of Review: This paper will review the current staging system for prostate adenocarcinoma patients, and will also review new information that can be combined with clinical and pathological staging in order to assess a patient's risk of success or failure of treatment.

Recent Findings: There has been significant stage migration of prostate cancer patients in the past 15 years, such that patients are currently being diagnosed younger, with lower clinical stages and serum prostate-specific antigen levels, and a lower risk of metastatic disease than previously. The incorporation of the results of extended prostate biopsy schemes, with stage, grade and serum prostate-specific antigen levels, improves the risk assessment of newly diagnosed prostate cancer patients.

A prospective comparison of 3 diagnostic methods to evaluate ejaculatory duct obstruction.

Purpose: Various diagnostic tests are available to evaluate patients with ejaculatory duct obstruction (EDO). However, the most accurate diagnostic technique, defined as the one that best predicts a successful outcome after ejaculatory duct resection, is unclear. We prospectively performed transrectal ultrasound (TRUS) and 3 other tests in men with EDO and determined their relative value in this diagnosis.

Purohit technique of vaginal hysterectomy: a new approach.

To ease intra-operative access to laterally at vaginal hysterectomy, we have developed the 'Purohit technique of vaginal hysterectomy' using a right angle forceps, electrocautery and 10 mm telescope with light source. A prospective study on consecutive 214 women with benign disease of the uterus without prolapse, including cases with relative contraindications (excluding endometriosis and uteri above 20 weeks size), demonstrated it to be easy, safe and effective. Colo-colic intussusception due to intestinal lipomatosis.

Lipomas of the small intestine are usually found incidentally; symptoms occur in less than one-third of affected patients, especially when lipomas are more than 2 cm in size. We report a 32-year-old man in whom intestinal lipomatosis resulted in colo-colic intussusception. These symptoms disappeared following operative resection of about 60 cm of ileum and ascending colon.

Laterality of symptomatic cystine calculi.

Objectives: To evaluate patterns of laterality in cystine stone formation to determine whether phenotypic expression of this genetic disease is distributed equally in each kidney. Cystinuria is a genetic defect that may result in the formation of recurrent cystine calculi. Significant nongenetic factors may play a role in the manifestation of this disease.

Imaging clinically localized prostate cancer.

At this time there is no highly sensitive and specific widespread radiographic test for local staging of prostate cancer. Future developments will likely require a combination of imaging modalities with utilization guided by risk-stratification models (Table 4). Staging data for all imaging tests discussed in this article are summarized in Tables 5 and 6.

Evaluation of the safety and efficacy of testicular biopsies in llamas.

Evaluation of the reproductive function of Lama glama is generally considered to be a challenging task due to the difficulty of obtaining representative semen samples. One method that has been proposed for evaluation of testicular function in these animals is histologic examination of testicular needle biopsies. This study was undertaken to examine the safety and efficacy of using needle biopsies to assess testicular function in this species.

Effects of testicular biopsy in clinically normal bulls.

Objective: To determine whether testicular needle biopsy is detrimental to testicular function in clinically normal bulls.

Animals: 6 mixed-breed mature bulls.

Cure of undifferentiated small cell carcinoma of the urinary bladder with M-VAC chemotherapy.

Small cell carcinoma (SCC) of the urinary bladder is a rare, aggressive malignancy with approximately 135 cases reported in the literature. Treatments have included chemotherapy, radical surgery, radiotherapy, and combinations of these. We present the apparent cure of a 73-year-old man who presented with clinical stage T2 SCC of the urinary bladder.

A comparison of surface and rectal temperatures between sheared and non-sheared alpacas (Lama pacos).

The objective of this research was to determine if whole-body shearing would effect gross thermoregulation in alpacas. Eight mature, intact male alpacas were randomly assigned to one of two groups and maintained in outdoor pastures with adequate artificial shade from June through August (summer climate) in east central Alabama, USA. Group one animals (N=4) were sheared to remove all fiber to within 2cm of their skin.

Successful long-term outcome using existing native cutaneous ureterostomy for renal transplant drainage.

Purpose: We report our long-term experience with a preexisting native cutaneous ureterostomy via an ipsilateral transplant ureteral native ureterostomy for transplant drainage without native nephrectomy.

Materials And Methods: Between 1993 and 1998, 5 patients without a usable bladder had undergone previously urinary diversion via cutaneous ureterostomy. All patients had a well functioning cutaneous ureterostomy for a mean plus or minus standard deviation of 18+/-12 years before renal transplantation.
