The ovary is one of the first organs to exhibit signs of aging, characterized by reduced tissue function, chronic inflammation, and fibrosis. Multinucleated giant cells (MNGCs), formed by macrophage fusion, typically occur in chronic immune pathologies, including infectious and non-infectious granulomas and the foreign body response , but are also observed in the aging ovary . The function and consequence of ovarian MNGCs remain unknown as their biological activity is highly context-dependent, and their large size has limited their isolation and analysis through technologies such as single-cell RNA sequencing.

Background: Ankle fracture is one of the most common lower limb fractures. Whilst immobilisation of the ankle can support and protect the fracture site during early healing, this also increases the risk of ankle weakness, stiffness, and residual pain. Rehabilitation aims to address the after-effects of this injury, to improve ankle function and quality of life.

Germline genomic sequencing is increasingly integrated into pediatric cancer care, with pathogenic cancer-predisposing variants identified among 5-18% of affected children and variants of uncertain significance (VUS) in up to 70%. Given the potential medical implications for children and their families, parents' psychosocial responses to learning results are important to understand. Parents of children with cancer who learned their children's germline pathogenic or VUS results following paired tumor and germline genomic sequencing described their cognitive and affective responses to results in an open-ended write-in question after disclosure (M = 10 months post-disclosure; range = 1-28).

The female reproductive system is one of the first to age in humans, resulting in infertility and endocrine disruptions. The aging ovary assumes a fibro-inflammatory milieu which negatively impacts gamete quantity and quality as well as ovulation. Here, we tested whether the systemic delivery of anti-inflammatory (Etanercept) or anti-fibrotic (Pirfenidone) drugs attenuates ovarian aging in mice.

Ovarian somatic cells are essential for reproductive function, but no existing models recapitulate the cellular heterogeneity or interactions within this compartment. We engineered a novel ovarian somatic organoid model by culturing a stroma-enriched fraction of mouse ovaries in scaffold-free agarose micromolds. Ovarian somatic organoids self-organized, maintained diverse cell populations, produced extracellular matrix, and secreted hormones.

Article Synopsis
  • - Acinar cystic transformation (ACT) is a rare, non-cancerous cystic lesion in the pancreas that's often confused with more serious conditions like intraductal papillary mucinous neoplasms (IPMNs).
  • - All known cases of ACT are benign and have not shown any signs of recurrence or cancer development, which complicates preoperative diagnosis.
  • - The Fukuoka guidelines assist in managing IPMNs, and in this case, a patient was misdiagnosed with a side-branch IPMN and underwent a Whipple's procedure; the report emphasizes the challenges in correct diagnosis and the tools available to healthcare professionals.

Nearly 85 % of emerging adults report using at least one social media site. Research suggests that viewing and internalizing unrealistic body ideals often displayed online may pose harmful effects on young people's body image. However, studies on the relationships between social media usage and body image have predominantly focused on women's drive for thinness.

Background Ankle fractures are very common injuries seen in an emergency setting. Initial management involves the application of below-knee plaster casts. At our local trauma meetings, we have observed that below-knee casts are often applied incorrectly which can result in suboptimal outcomes for patients and increase the burden on plaster room services if re-application is required.

Article Synopsis
  • About one-third of older adults aged 65 and older fall each year, prompting the need for community-wide interventions that address the broader factors contributing to these falls, such as environmental hazards and lack of physical activity.
  • These interventions may include policies for increased vitamin D intake, reducing hazards in homes and communities, and promoting public health programs that encourage exercise among older adults.
  • The study aimed to review evidence on the effectiveness of these population-based interventions by analyzing randomized controlled trials and other relevant studies, focusing on strategies that benefit entire communities rather than just individuals at high risk.

Article Synopsis
  • Global storm-resolving models (GSRMs) have gained popularity for their detailed representation of global climate, but assessing their differences in simulating atmospheric formations poses challenges.
  • To overcome this, the study introduces methods that analyze distributional distances through nonlinear dimensionality reduction and vector quantization, allowing for a more objective comparison of model outputs.
  • The findings indicate that only six out of nine models exhibit similar representations of atmospheric dynamics, while also revealing signs of the convective response to global warming, paving the way for better evaluation of high-resolution simulation data.
