Nicotine is acknowledged as the key addictive compound of tobacco. Varenicline (Champix or Chantix), mainly acting as a partial agonist at the α4β2 nicotinic receptor, is an approved smoking cessation pharmacotherapy, although with efficacy limited to a portion of smokers. Smokers differ in the motives that drive their drug seeking and Varenicline might be more efficient in some groups more than others.

Inflexible behavior is a hallmark of several decision-making-related disorders such as ADHD and addiction. As in humans, a subset of healthy rats makes poor decisions and prefers immediate larger rewards despite suffering large losses in a rat gambling task (RGT). They also display a combination of traits reminiscent of addiction, notably inflexible behavior and perseverative responses.

Article Synopsis
  • High-frequency cocaine intake is linked to increased drug-seeking behavior in both humans and animals, but the underlying neurobiology is not fully understood.
  • Key brain structures like the medial prefrontal cortex (mPFC), basolateral amygdala (BLA), and nucleus accumbens (NAc) play roles in cocaine seeking, and changes in the immediate early gene c-Fos were studied to explore their contributions.
  • The research found that high-frequency cocaine self-administration in rats led to enhanced c-Fos activity in specific mPFC regions, and manipulating these areas affected drug-seeking behavior differently based on the self-administration frequency, suggesting individual variations might influence vulnerability to cocaine addiction.
