Lymph node tissue of eight cases of Hodgkin's disease of all different subtypes was studied with an immunoperoxidase technique for the detection of immunoglobulin G (IgG), J chain, kappa and lambda light chains, and alpha-1-antitrypsin in different types of Sternberg-Reed cells. It was found that L&H type Sternberg-Reed cells of the nodular lymphocyte predominance type contained IgG, J chain, and one type of light chain per individual cell. It is concluded that these findings indicate that L&H type Sternberg-Reed cells produce IgG and, consequently, are B immunoblasts.
View Article and Find Full Text PDFThe histopathologic classification of Hodgkin's disease is discussed in relation to age and sex of 278 children younger than 15 years of age, whose lymph node biopsies were diagnosed between 1963 and 1978 at the Lymph Node Registry in Kiel. The distribution of the histologic subtypes was not essentially different from that in adult cases, with the exception of a lower incidence of the lymphocyte depletion type in children. The youngest patient was 22 months old.
View Article and Find Full Text PDFLymphoid tissue of 51 patients with Hodgkin's disease was studied with immunohistological, enzyme histochemical and rosetting techniques for the detection of B and T cells in frozen sections. In lymph nodes of patients with lymphocyte predominance type of Hodgkin's disease, the majority of the lymphocytes in the involved areas were normal B lymphocytes of polyclonal origin. This was also true for nodular sclerosis cases with a predominance of lymphocytes.

The age and sex distribution and the localization of excised lymph nodes from 206 patients with a nodular variant of Hodgkin's disease with lymphocytic predominance, called "nodular paragranuloma", are presented and compared with data on other types of Hodgkin's disease. The age curve of nodular paragranuloma showed a peak in the fourth decade, which was clearly separated from the peak in the third decade exhibited by the nodular sclerosis type of Hodgkin's disease and from the peak in the sixth and seventh decades of the mixed cellularity type. The peak in the age curve of nodular paragranuloma resulted from the high frequency in men in the fourth decade; the female age curve had no peaks.

The histological features of nodular paragranuloma are compared with those of progressively transformed germinal centres, which are enlarged follicles showing a predominance of small lymphocytes and some residual germinal centre cells. Progressively transformed germinal centres are sometimes found in nonspecific lymphadenitis (reactive hyperplasia).

Ultrastructural and immunohistologic findings in a nodular variant of Hodgkin's disease with lymphocytic predominance, called nodular paragranuloma, are presented and compared with those in so-called progressively transformed germinal centers. These are large follicles with numerous lymphocytes which can be found not only in nonspecific lymphadenitis, but also in lymph nodes from patients with nodular paragranuloma. The immunoperoxidase technique was applied on paraffin sections to detect intracytoplasmic immunoglobulin and lysozyme.

Fifteen primary intracranial reticulum cell sarcomas and five cases with an additional solitary extracranial tumor mass have been studied. For comparison, seven extracranial malignant non-Hodgkin lymphomas and normal lymphoid tissue were included. The methods used on formalin-fixed paraffin-embedded tissue sections were an immunoperoxidase technique for the demonstration of intracellular immunoglobulins, microglial staining, Gomori's reticulin, methylgreen-pyronin, Giemsa, diastase resistant PAS, Mallory's PTAH and H&E.

Phagocytosis by Sternberg-Reed cells is a rare phenomenon. A case of nodular sclerosis type of Hodgkin's disease with large numbers of apparently vital lymphocytes in the cytoplasm of the Sternberg-Reed cells is described in this paper. The question whether this feature is the result of phagocytosis or emperipolesis is discussed.

The presence of IgG in some Reed-Sternberg (R-S) cells was confirmed, and in addition serial sections and a double staining technique revealed that these cells also contained both kappa and lambda light chains. Furthermore two serum proteins, human serum albumin and alpha-1-antitrypsin, were demonstrated in the same positive R-S cells.
View Article and Find Full Text PDFSkin biopsies and a lymphnode of three children with infantile Histiocytosis-X (Letter-Siwe Disease) were studied with enzymehistochemical and sheep-erythrocyte rosetting techniques. The majority of cells making up the infiltrates of skin and lymphnode showed rather weak acid phosphatase and nonspecific esterase activity but considerable leucyl-beta-naphtylamidase activity. Sheep-erythrocyte rosetting techniques performed on frozen sections indicated the presence of receptors for the Fc fragment of IgG, but no receptors for C3 could be demonstrated.
