Lampbrush chromosomes, with their unusually high rate of nascent RNA synthesis, provide a valuable model for studying mechanisms of global transcriptome up-regulation. Here, we obtained a whole-genomic profile of transcription along the entire length of all lampbrush chromosomes in the chicken karyotype. With nuclear RNA-seq, we obtained information about a wider set of transcripts, including long non-coding RNAs retained in the nucleus and stable intronic sequence RNAs.

Analyses of ancient DNA typically involve sequencing the surviving short oligonucleotides and aligning to genome assemblies from related, modern species. Here, we report that skin from a female woolly mammoth (†Mammuthus primigenius) that died 52,000 years ago retained its ancient genome architecture. We use PaleoHi-C to map chromatin contacts and assemble its genome, yielding 28 chromosome-length scaffolds.

A number of species have recently recovered from near-extinction. Although these species have avoided the immediate extinction threat, their long-term viability remains precarious due to the potential genetic consequences of population declines, which are poorly understood on a timescale beyond a few generations. Woolly mammoths (Mammuthus primigenius) became isolated on Wrangel Island around 10,000 years ago and persisted for over 200 generations before becoming extinct around 4,000 years ago.

Article Synopsis
  • Scientists studied 23 woolly mammoth genomes to learn how these creatures adapted over time, even looking at one that was 700,000 years old.
  • They found that the woolly mammoth already had many important genes for things like hair, fat storage, and its immune system when it first appeared.
  • The research shows that different genes helped the woolly mammoth change during its long existence, including some that affected its size and ear shape in later years.

Background: Karyotype abnormalities are frequent in immortalized continuous cell lines either transformed or derived from primary tumors. Chromosomal rearrangements can cause dramatic changes in gene expression and affect cellular phenotype and behavior during in vitro culture. Structural variations of chromosomes in many continuous mammalian cell lines are well documented, but chromosome aberrations in cell lines from other vertebrate models often remain understudied.

Article Synopsis
  • The grey wolf was the first animal to be domesticated into dogs and lived through the last Ice Age when many other animals went extinct.
  • Scientists studied 72 ancient wolf genomes from different places to understand how wolves were connected and how they evolved over 100,000 years.
  • They found that dogs are more closely related to ancient wolves from eastern Asia, but some dogs in the Near East and Africa share ancestors with different wolves, which means there might have been several ways dogs were domesticated.

The cave lion is an extinct felid that was widespread across the Holarctic throughout the Late Pleistocene. Its closest extant relative is the lion (Panthera leo), but the timing of the divergence between these two taxa, as well as their taxonomic ranking are contentious. In this study we analyse 31 mitochondrial genome sequences from cave lion individuals that, through a combination of C and genetic tip dating, are estimated to be from dates extending well into the mid-Pleistocene.

The first description of well-preserved fur of one male and one female cubs of the fossil cave lion Panthera spelaea (finds of 2017-2018, Semyuelyakh River, Yakutia, Russia) is presented in comparison with the fur of two cubs (a male and a female) of the extant African lion P. leo and adult lions of both species: the color, differentiation, configuration, microstructure, and internal design of hair of different categories are considered. The coat of the cave lion is similar but not identical to that of the African lion, because it contains a high thick undercoat of spiral-shaped thick and airy down hair.

Ancient remains found in permafrost represent a rare opportunity to study past ecosystems. Here, we present an exceptionally well-preserved ancient bird carcass found in the Siberian permafrost, along with a radiocarbon date and a reconstruction of its complete mitochondrial genome. The carcass was radiocarbon dated to approximately 44-49 ka BP, and was genetically identified as a female horned lark.

Ancient DNA provides a powerful means to investigate the timing, rate and extent of population declines caused by extrinsic factors, such as past climate change and human activities. One species probably affected by both these factors is the arctic fox, which had a large distribution during the last glaciation that subsequently contracted at the start of the Holocene. More recently, the arctic fox population in Scandinavia went through a demographic bottleneck owing to human persecution.

The 28,000-year-old remains of a woolly mammoth, named 'Yuka', were found in Siberian permafrost. Here we recovered the less-damaged nucleus-like structures from the remains and visualised their dynamics in living mouse oocytes after nuclear transfer. Proteomic analyses demonstrated the presence of nuclear components in the remains.

We experimentally study the structure and dynamics of magnetic domains in synthetic antiferromagnets based on Co/Ru/Co films. Dramatic effects arise from the interaction among the topological defects comprising the dual domain walls in these structures. Under applied magnetic fields, the dual domain walls propagate following the dynamics of bi-meronic (bi-vortex/bi-antivortex) topological defects built in the walls.

Based on the pathopsychological analysis, the author suggests a hypothesis of integrative basis of negative disorders of cognition in schizophrenia. The disorders are proposed to be related to the infringement of the general principle of CNS functioning - the domination of the process unifying cognitive, subjective-psychological, neurophysiological components of individuality.

The growth, composition and structure of sandwich structures (Fe-rich layer/Si-rich layer/Fe-rich silicide layer) grown on a Si(111) surface were studied by a few complementary microscopic and spectroscopic techniques with high spatial resolution. Intermixing at the Fe/Si and Si/Fe interfaces is demonstrated. Fe-rich layers grown directly on the Si(111) surface are crystalline and have abrupt but rough interfaces at both sides.

The paper presents the first morphological description of the internal organs of a frozen corpse of the steppe bison Bison priscus (Bojanus, 1827) from the Holocene of northern Yakutia. Necropsy revealed that most of the internal organs, including the brain, heart with the main vessels, and reproductive system were well preserved. It demonstrated that the anatomy of this bison was close to that of the genera Bos and Bison.

We have discovered that films of carbon single wall nanotubes (SWNTs) make excellent back contacts to CdTe devices without any modification to the CdTe surface. Efficiencies of SWNT-contacted devices are slightly higher than otherwise identical devices formed with standard Au/Cu back contacts. The SWNT layer is thermally stable and easily applied with a spray process, and SWNT-contacted devices show no signs of degradation during accelerated life testing.
