In the present study, 28 hemophiliacs substituted continuously and 5 hemophiliacs who had received almost no blood products were investigated. Cells of OKT 3+, OKT 4+, and OKT 8+ subsets were counted. Percoll separated fractions of peripheral blood mononuclear cells were examined by morphological criteria and were tested for NK cell activity.

Eight similar reports in the medical literature suggested it to be a rare case of intolerance to hydrochlorothiazide. The lymphocyte transformation test proved an allergic genesis: there was significant stimulation of patient lymphocytes by active agent/metabolite serum of hydrochlorothiazide but not triamterene.

(Other markers, such as ouabain- or methothrexate-resistance and alanine independence, in other L5178Y mouse lymphoma cells were also examined, but our criteria for the acceptability of data or the paucity of data considerably reduced the value of these mutagenesis test systems to this study.) The biochemical basis for the L5178Y/TK+/- assay depends on the ability of TK-competent cells to phosphorylate 5-bromo-2'-deoxyuridine or trifluorothymidine.

Chloropropanol was tested over a dose range of 527-167,250 micrograms/plate in the Salmonella/mammalian microsome mutagenicity assay. A dose dependent mutagenic response was observed in strains TA 1535 and TA 100.

The diuretic effects of bemetizide and hydrochlorothiazide were qualitatively equal, though after higher doses bemetizide showed greater maximal, statistically significant effects on the excretion of sodium, chloride and urine volume (relative efficacy). A trend to increased elimination of potassium was detectable.

Eleven metals were examined for their potential to induce forward mutations at the thymidine kinase locus in L5178Y mouse lymphoma cells. The materials tested included AlCl3, CdSO4, HgCl2, K2CrO4, K2Cr2O7, MgCl2, MnCl2, NaAsO2, Na2HAsO4, NaCl, and Pb(NO3)2. Strong positive responses at survivals greater than 10% were observed with CdSO4, K2CrO4, K2Cr2O7, and MnCl2.

The role of colostrum and milk in the transmission of the bovine leukemia virus (BLV) was examined by monitoring the development of BLV infection in calves that were fed since birth on colostrum and milk from their BLV-positive dams and then reared in complete or partial isolation from infected cattle. Twenty-one of the 25 calves raised in complete isolation remained negative for BLV until the last evaluation. At this time, 14 calves were older than 28 months of age, and seven were 16 to 20 months old.

The battery consisted of the Salmonella/mammalian microsome assay, the L5178Y mouse lymphoma assay, and unscheduled DNA synthesis using W138 cells. The mutagenic potential of the compounds ranged from strongly mutagenic to non-mutagenic; glycidol exhibited the greatest activity.

Data are presented from tests of 77 compounds representing a wide range of chemical classes which demonstrate the efficiency of the E coli microsuspension assay as both a qualitative and quantitative screen of DNA-modifying activity. Furthermore, the use of a battery of indicator strains lacking different repair systems offers the advantage of providing preliminary information concerning the mechanism of DNA damage induction by a test agent.

Patients who still have symptoms after years of such therapy are referred to us for surgical treatment. Failure to obtain symptomatic relief after scalenotomy alone [8] caused us to abandon this treatment.

Of 7 cattle (group 1) exposed beginning in July and August (midsummer) of 1976, 4 were infected by October, and all 7 by November (4 months' exposure).

The use of early-passage indicator bovine embryonic spleen (BESP) cells and their pretreatment with diethylamino-ethyl-dextran (DEAE-D) was essential for optimal sensitivity. Polybrene was less effective than DEAE-D.

Only 3 of the 17 isolated cattle became infected during 25-29 months of observation. During the same period of time, all 17 cattle raised in contact with BLV-positive animals developed BLV infection.

One should still attempt conservative management of the lesion with aspiration therapy, tube thorocostomy and the adminstration of medium length triglyceride fatty acid diets. However, if this is not successful within two to three weeks, one should not wait for disastrous nutritional degeneration before recommending this form of surgical intervention.

The release of infectious BLV and spontaneous incorporation of [3H]thymidine were not properties of density gradient-fractionated leukocytes from a BLV-free cow. When leukocytes from BLV-infected cattle were fractionated, B lymphocytes which spontaneously incorporated [3H]thymidine could be separated as a distinct subpopulation from B lymphocytes which replicated infectious BLV.

Direct comparisons of the percentages of S-Ig-bearing cells and EAC rosette-forming cells in both infected and BLV-free animals showed no evidence for the presence of a substantial population bearing one surface marker but not the other. The data showed that cells with surface markers characteristic of B lymphocytes are responsible for most of the increase in peripheral blood lymphocytes which may accompany BLV infection.
