Metastatic uveal melanoma is a rare disease with a poor prognosis. Usual treatments have not proven effective. Tebentafusp, a bispecific protein targeting melanoma cells and T lymphocytes, is the first approved treatment with a proven survival benefit in a randomized clinical.

Article Synopsis
  • The study examines the long-term effectiveness of immune checkpoint inhibitor (ICI) rechallenge in patients with advanced melanoma who previously responded to ICI treatment.
  • A total of 85 patients participated, with a notable 54% showing a positive response to the rechallenge, including 35% achieving a complete response.
  • However, safety concerns emerged, as 27% experienced adverse events, and those who underwent other treatments between ICI courses had lower response rates and shorter progression-free survival.

Cemiplimab, a human monoclonal antibody directed against PD-1, has provided more options in the treatment of locally advanced or metastatic cutaneous squamous-cell carcinoma at an unresectable state. Immune checkpoint inhibitors can induce several unfavorable reactions generally referred to as immune-related adverse effects. Cytokine-release syndrome is an immune-related adverse event that is infrequent and not well known.

Article Synopsis
  • Cemiplimab has shown promise in real-world use for locally advanced and metastatic cutaneous squamous-cell carcinomas (CSCCs), with a best overall response rate of 50.4% among treated patients.
  • There was a median progression-free survival (PFS) of 7.9 months, and one-year overall survival (OS) was 73% for patients with better performance status (PS < 2).
  • Although some patients experienced severe adverse events, the overall safety and efficacy indicate cemiplimab may be a beneficial treatment option, especially for immunocompromised individuals.

In BRAF wild type advanced melanoma, immune checkpoint blockers such as anti-PD1 (anti-programmed cell death 1) are usually continued beyond progression for a hypothetical rare further response. Chemotherapy as a second-line option is considered ineffective by many practitioners based on historical data. Continuing anti-PD1 beyond progression has a high health-economic impact and is not recommended by the FDA.

Background: Anti-PD1 antibodies have revolutionized the management of patients with advanced melanoma. In clinical trials, the efficacy of nivolumab is being tested in selected populations of patients.

Objectives: The aim of this study was to analyse the efficacy and safety of nivolumab in patients with advanced melanoma under real-life conditions.

For melanoma patients, surgery is a standard treatment for locoregional skin metastasis (LSM). To assess the frequency and risk factors for positive margins after excision of LSM and their impact on patient overall survival (OS) and progression-free survival (PFS). A monocentric, retrospective observational study was performed including 87 patients with LSM who had undergone surgical excision.

Immunotherapy for melanoma includes adoptive cell therapy with autologous tumor-infiltrating lymphocytes (TILs). This monocenter retrospective study was undertaken to evaluate the efficacy and safety of this treatment of patients with advanced melanoma. All advanced melanoma patients treated with TILs using the same TIL expansion methodology and same treatment interleukin-2 (IL-2) regimen between 2009 and 2012 were included.

Purpose: The dermatological toxicity of cancer treatments is frequent and sometimes debilitating. Its reference classification, the NCI-CTCAE (National Cancer Institute-Common Terminology Criteria for Adverse Events), is sometimes difficult to use and does not include yet the newest toxicities. Our objective was to create a guide, TOXICAN, based on the CTCAE, which is easy to use in everyday practice and which facilitates the recognition and grading of these dermatological toxicities.

Article Synopsis
  • Nivolumab has a 40% response rate in metastatic melanoma, and the study aimed to identify blood biomarkers that predict survival for patients treated with it.
  • Researchers evaluated 9 potential blood biomarkers from advanced melanoma patients over a 3-year period and used statistical analyses to find associations with overall and progression-free survival.
  • Key findings indicated that higher levels of monocyte count, leukocyte/lymphocyte ratio, and neutrophil/lymphocyte ratio were linked to lower overall survival and progression-free survival, suggesting these markers could be helpful in forecasting patient responses to anti-PD1 therapy.

Few satisfactory treatment options are available for widespread areas affected by multiple actinic keratoses (AKs). Our primary objective was to assess the response rate to weekly 5-fluorouracil (5-FU) chemowraps on widespread AK lesions, and secondarily to assess tolerability, the percentage of patients with recurrence and time to recurrence, the response rate for patients with associated Bowen's disease (BD), and the percentage of squamous cell carcinomas (SCCs) identified after treatment. We conducted an open study which included all the patients who had been treated with weekly 5-FU chemowraps in our department over the course of five years for areas of widespread AKs.

Data on BRAF, NRAS and KIT mutations are scarce in patients with vulvo-vaginal melanomas and are associated with important therapeutic issues. We investigated their prevalence in a cohort of patients with female lower genital tract melanomas between 2003 and 2017. Of the 22 patients, 5 (22.

Background: Immune checkpoint blockade therapy (ICBT) for the treatment of melanoma has led to an important improvement of overall survival in advanced stage patients. However, secondary cutaneous maculopapular eruptions (CMPEs) are frequent and remain poorly characterized.

Methods: We performed a retrospective analysis of melanoma patients from our institution who developed CMPEs during ICBT.
