SARS-CoV-2 can induce vascular dysfunction and thrombotic events in patients with severe COVID-19; however, the cellular and molecular mechanisms behind these effects remain largely unknown. In this study, we used a combination of experimental and in silico approaches to investigate the role of PC in vascular and thrombotic events in COVID-19. Single-cell RNA-sequencing data from patients with COVID-19 and healthy subjects were obtained from the publicly available Gene Expression Omnibus (GEO) repository.

Article Synopsis
  • Chikungunya virus (CHIKV) can lead to severe diseases, including chronic arthritis and, in rare cases, neurological issues and death, primarily transmitted by mosquitoes.
  • A comprehensive study revealed that deaths from CHIKV are linked to multi-organ infections, serious brain damage, and higher levels of inflammation in the body compared to survivors.
  • The research also found that CHIKV infection disrupts the blood-brain barrier, leading to increased permeability and changes in protein expression, enhancing our understanding of CHIK pathophysiology and fatal outcomes.

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the agent of a major global outbreak of respiratory tract disease known as Coronavirus Disease 2019 (COVID-19). SARS-CoV-2 infects mainly lungs and may cause several immune-related complications, such as lymphocytopenia and cytokine storm, which are associated with the severity of the disease and predict mortality. The mechanism by which SARS-CoV-2 infection may result in immune system dysfunction is still not fully understood.

Mayaro virus (MAYV) is an arthropod-borne virus (arbovirus) belonging to the family Togaviridae, genus Alphavirus. In recent years, the geographic distribution of MAYV may have expanded north from South and Central America into the Caribbean Islands. Although Haemagogus janthinomys is considered the main vector for MAYV, the virus has also been isolated from other mosquitoes, including Aedes aegypti, a widespread species that serves as the main vector for highly epidemic viruses.

The duration and protectiveness of antibodies against SARS-CoV-2 in infected subjects are still uncertain; nonetheless, anti-S-specific antibodies can contribute to protective immunity against new infections. It has been described that the level of antibodies produced in COVID-19 is related to the severity of symptoms, and the majority of the humoral response studies have been conducted in hospitalized patients who have been, then, followed over time. However, about 80% of SARS-CoV-2 infections in unvaccinated people are mild to asymptomatic, and this percentage reaches more than 95% in vaccinated individuals.

The COVID-19 (Coronavirus Disease 2019), caused by the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), severely affects mainly individuals with pre-existing comorbidities. Here our aim was to correlate the mTOR (mammalian/mechanistic Target of Rapamycin) and autophagy pathways with the disease severity. Through western blotting and RNA analysis, we found increased mTOR signaling and suppression of genes related to autophagy, lysosome, and vesicle fusion in Vero E6 cells infected with SARS-CoV-2 as well as in transcriptomic data mining of bronchoalveolar epithelial cells from severe COVID-19 patients.

Article Synopsis
  • The Italian Research Group for Gastric Cancer (GIRCG) noted in 2013 that laparoscopic or robotic surgeries should primarily be used for early gastric cancer (EGC), with no further guidelines available at that time.
  • A recent study surveyed 46 out of 100 expert Italian upper gastrointestinal surgeons on their use of minimally invasive techniques for gastric cancer treatment, finding a similar preference for these methods in both early and advanced gastric cancer cases.
  • The results indicated that as the number of surgical procedures performed at a center increased, so did the likelihood of utilizing minimally invasive approaches, suggesting a shift in clinical practice beyond current national guidelines.

Although increasing evidence confirms neuropsychiatric manifestations associated mainly with severe COVID-19 infection, long-term neuropsychiatric dysfunction (recently characterized as part of "long COVID-19" syndrome) has been frequently observed after mild infection. We show the spectrum of cerebral impact of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, ranging from long-term alterations in mildly infected individuals (orbitofrontal cortical atrophy, neurocognitive impairment, excessive fatigue and anxiety symptoms) to severe acute damage confirmed in brain tissue samples extracted from the orbitofrontal region (via endonasal transethmoidal access) from individuals who died of COVID-19. In an independent cohort of 26 individuals who died of COVID-19, we used histopathological signs of brain damage as a guide for possible SARS-CoV-2 brain infection and found that among the 5 individuals who exhibited those signs, all of them had genetic material of the virus in the brain.

Article Synopsis
  • The COVID-19 pandemic has highlighted the need for effective treatments, prompting research into new antiviral compounds.
  • A novel class of steroidal compounds, LNB167 and LNB169, shows promise against SARS-CoV-2, reducing viral replication in human cell lines.
  • However, while these compounds exhibit in vitro effectiveness, they failed to provide protection in live animal models, suggesting that their mechanism of action is complex and not directly translatable to all biological systems.

Article Synopsis
  • SARS-CoV-2, the virus causing the COVID-19 pandemic, interacts with the human protein Proliferating Cell Nuclear Antigen (PCNA), which is important for DNA repair and replication.
  • The study used several methods to confirm this interaction, finding that in infected cells, PCNA shifts from the nucleus to the cytoplasm and has increased expression levels, indicating DNA damage.
  • Targeting the PCNA-M protein interaction could be a potential therapeutic strategy for COVID-19, as inhibiting this process reduced viral plaque formation in experiments.

  • Previous studies show that people who have had COVID-19 develop strong antibody responses after vaccination.
  • In a study of 21 vaccinated individuals, researchers compared antibody levels before and after immunization between those who had recovered from COVID-19 and those who had never been infected.
  • The findings revealed that while naïve individuals showed different antibody responses after two vaccine doses, recovered individuals did not see significant increases after the second dose, indicating that just one dose may be enough for those previously infected to generate protective antibody responses.
