Environmental hazardous material polychlorinated biphenyl (PCB) exposure is associated with vascular endothelial dysfunction, which may increase the risk of cardiovascular diseases and cancer metastasis. Our previous studies illustrated the cytotoxic, antiproliferative, and genotoxic effects of a synthetic, quinone-type, highly reactive metabolite of PCB, 2,3,5-trichloro-6-phenyl-[1,4]benzoquinone (PCB29-pQ). Here, we used it as the model compound to investigate its effects on vascular endothelial integrity and permeability.

Diphenyl difluoroketone (EF24), a curcumin analog, exhibits potent anti-tumor activities by arresting cell cycle and inducing apoptosis. However, the efficacy and modes of action of EF24 on melanoma metastasis remain elusive. In this study, we found that at non-cytotoxic concentrations, EF24 suppressed cell motility and epithelial-to-mesenchymal Transition (EMT) of melanoma cell lines, Lu1205 and A375.

Article Synopsis
  • Melanoma cell migration through vascular endothelial cells is crucial for tumor metastasis, and the protein RacGAP1 plays a significant role in this process.
  • Knocking down RacGAP1 or using a mutant version hinders melanoma cell movement and disrupts cell junctions.
  • RacGAP1 activation leads to the formation of focal adhesions and cytoskeletal changes, affecting the endothelial barrier and facilitating melanoma cell transmigration.
