Article Synopsis
  • Salivary gland adenoid cystic carcinoma (ACC), mucoepidermoid carcinoma (MEC), and oral squamous cell carcinoma (OSCC) are types of cancer in the head and neck, with previous treatments like immune checkpoint inhibitors failing in ACC.
  • The study investigated the effect of Gipie (CCDC88B), a protein that activates immune cells, by silencing it in cancer cell co-culture models and using various imaging and proteomic techniques.
  • Results showed that silencing Gipie in ACC cells led to increased cell death and activation of immune cells, suggesting that Gipie hinders anti-tumor immune responses in ACC but had little effect on other cancer types.

Objectives: Signal transducer and activator of transcription 3 (STAT3) is one of the key proliferation mechanism-related proteins that helps in oral squamous cell carcinoma (OSCC) progression. Immune evasion by STAT3 is mediated by the JAK2/STAT3/PDL1 signaling axis. Based on previous findings, we hypothesized that STAT3-binding partners participate in the inhibition of anti-tumor activity in OSCC.

Aims: Ochratoxin A (OTA) has been reported to exhibit nephrotoxicity through induction of cell redox homeostasis perturbation, mitochondrial hyperpolarization and depolarization, protein synthesis inhibition, apoptosis, etc. In the present examination, the protective efficiency of novel synthesized molecule, N-acetyl-L-Tryptophan glucoside (NATG) towards OTA prompted toxicity was evaluated using Human Embryonic Kidney (HEK-293) cells.

Main Methods & Key Findings: The cells were treated with NATG (0-200 μg/ml) before OTA treatment (0-20 μg/ml) the and protection efficiency of NATG was evaluated using MTT and SRB assay.
