Article Synopsis
  • - Cigarette smoke (CS) negatively impacts lung cancer progression and patient outcomes, with current research focusing on how it affects macrophage behavior in the lungs of smokers.
  • - The study used murine macrophages and cigarette smoke extract (CSE) to show that CS promotes polarization of macrophages towards an M2 phenotype, which are known to be more resistant to the harmful effects of CS.
  • - The results suggest that CS encourages a predominance of M2 macrophages in the lungs of smokers, creating an anti-inflammatory and potentially immunosuppressive environment that worsens lung cancer conditions.

Background: Microsatellite instability high (MSI-H) and/or mismatch repair deficient (dMMR) status is the strongest predictive factor for immune checkpoint inhibitors (ICIs) benefit in patients with metastatic gastroesophageal cancer (mGC). Primary resistance to ICIs is a relevant issue, but prognostic and predictive factors are lacking.

Materials And Methods: In this multinational, retrospective cohort of patients with MSI-H/dMMR mGC treated with ICIs without chemotherapy we collected baseline laboratory values to establish the prognostic nutritional index (PNI).

To recognize patients at high risk of refractory disease, the identification of novel prognostic parameters improving stratification of newly diagnosed Hodgkin Lymphoma (HL) is still needed. This study investigates the potential value of metabolic and texture features, extracted from baseline 18F-FDG Positron Emission Tomography/Computed Tomography (PET) and Contrast-Enhanced Computed Tomography scan (CECT), together with clinical data, in predicting first-line therapy refractoriness (R) of classical HL (cHL) with mediastinal bulk involvement. We reviewed 69 cHL patients who underwent staging PET and CECT.

Article Synopsis
  • Myeloablative T cell depleted hematopoietic cell transplantation (HCT) leads to lower rates of acute and chronic graft versus host disease (GVHD) and can allow patients to respond to vaccinations post-transplant.
  • A study of 251 patients showed that 30.7% were vaccinated, with varying success rates in generating protective antibodies for diseases like hepatitis and pneumococcus.
  • Increased counts of specific T and B cell subsets may help predict which patients will have a better response to vaccinations after CD34-selected HCT, despite no significant differences noted for those who received rituximab treatment.

Malignant pleural mesothelioma (MPM) is an aggressive cancer associated with asbestos exposure. MPM pathogenesis has been related both to oxidative stress, evoked by and in response to asbestos fibers exposure, and epithelial mesenchymal transition (EMT), an event induced by oxidative stress itself and related to cancer proliferation and metastasis. Asbestos-related primary oxidative damage is counteracted in the lungs by various redox-sensitive factors, often hyperactivated in some cancers.

Lung cancer (LC) represents the leading cause of cancer incidence and mortality worldwide. LC onset is strongly related to genetic mutations and environmental interactions, such as tobacco smoking, or pathological conditions, such as chronic inflammation. Despite advancement in knowledge of the molecular mechanisms involved in LC, this tumor is still characterized by an unfavorable prognosis, and the current therapeutic options are unsatisfactory.

  • Researchers studied COVID-19 patients in Brazil to identify molecular biomarkers linked to disease severity, focusing on inflammatory mediators in patients compared to healthy controls.
  • They found that critically ill patients showed elevated serum levels of multiple inflammatory mediators, including HMGB1, which is associated with worse outcomes.
  • The study suggests that high levels of HMGB1 (over 125.4 ng/ml) could serve as a useful biomarker for predicting severe cases and increased risk of death in COVID-19 patients.
