The study was designed to investigate contribution of glucocorticoid hormones and capsaicin-sensitive neurons to gastroprotective effect of ischemic preconditioning against injury induced by 3.5 h gastric ischemia-reperfusion in rats. The contribution of glucocorticoids was investigated by an inhibition of corticosterone synthesis with metyrapone and adrenalectomy followed by corticosterone replacement.
View Article and Find Full Text PDFOne week after injection of streptozotocin (60 mg/kg intravenously), rats developed diabetes associated with a significant increase of gastric mucosa sensitivity to the ulcerogenic effect of indomethacin (35 mg/kg subcutaneously). Since potentiation of the ulcerogenic effect of indomethacin was observed only in rats subjected to fasting before drug injection, we hypothesize that this effect was caused by a drop of high glucose level in the blood after fasting.

To estimate the role of glucocorticoids adrenalectomy followed by appropriate corticosterone replacement and inhibition of corticosterone synthesis by metyrapone were used. Prostaglandin synthesis was inhibited by indomethacin.

Gastric ischemic preconditioning is one of the most effective approaches to attenuate the gastric mucosal injury caused by severe ischemia-reperfusion. In the present study we investigated whether glucocorticoid hormones participate in a realization of gastroprotective effect of ischemic preconditioning. Rats were exposed to prolonged gastric ischemia-reperfusion (30 min occlusion of celiac artery followed by 3 h of reperfusion) alone or with brief preliminary ischemic-reperfusion preconditioning (two 5 min episodes of occlusion of celiac artery followed by 10 min reperfusion).

The investigation concerned glucocorticoid hormones participation in gastroprotection during ischemia and ischemia-reperfusion in rats. Gastric ischemia was performed by occlusion of celiac artery in anaesthetized rats. The stomach was exposed to brief ischemia as well as brief or prolonged ischemia-reperfusion.
